Dictamnus dasycarpus Turcz. is the drying root of Dictamus dasyccarpus Turcz. and D.angustifolius G. Doc, and as the traditional medicine for dermatosis, heat-clearing and damp-drying drug and wind-dispelling drug. Many pharmacological activitities compounds exist, for example, detoxication. alkaloids, lactone, sesquiterpenes and so on. In the paper, the process of extraction of the bioactive components is researched, a new method for isolating and purifying the chemical composition preliminary established, and the activity of these compounds are discussed.The process of extraction for mainly constituents from Dictammis dasycarpus Turcz. is tested, ultrasonic extraction, reflux and warm immersion are used and compared, then the content of consituents is examined by high-performance liquid chromatography (HPLC), the results showed that the reflux is the best method for extraction, because it would not destroy the activities of compounds and is efficient. Then the ethanol concentration, solid-liquid ration, extraction time and extraction times are reviewed, and the influences of them are investigated, lastly, the best process is obtained from the orthogonal test, which is 15 times 80% ethanol extract 3 times and lhour every time at the ratio of material to liquid.High performance centrifugal partition Chromatography (HPCPC) method for the separation and purification of chemical constituents is used. In the paper, solvent system election, flow rate and rotation speed are experiment, three kinds of compounds are successfully separated from Dictammis dasycarpus Turcz. To separate Fraxinellone, use the solvent system composing of hexane-ethylacetate-ethanol-water (1:1:1: 1, v/v/v/v), the lower phase as the mobile phase and the separate conditions are flow rate is 1.5mL/min and the revolution speed at 1000 rpm; To separate Dictaminine and Obacunone, use the solvent system composing of hexane-ethylacetate-ethanol-water (1:1:1:1, v/v/v/v), the lower phase as the mobile phase and the separate conditions are flow rate is 1.3 mL/min and the revolution speed at 1200 rpm. The fractions are identified by high-performance liquid chromatograph (HPLC) for their retention times with the standards, and the purities of these compounds are above 98%. ESI-MSn are applied to examine the structure information and identify these compounds.DPPH method is applied to study the antioxidant activity of Dictammis dasycarpus Turcz.for the first time, the different extraction solvents and extraction methods are tested. The results show that the extract of Dictammis dasycarpus Turcz. have a better antioxidant activity, the best method is reflux and the best solvent is ethanol for the higher scavenging rates.The fungicidal activity are also researched, firstly, the samples of different extract solvents are tested, ethanol, acetoacetate, n-butanol, petroleum, water, secondly, the fungicidal activity of the extract and the fractions and them for the different bacteria, Staphylococcus aureus, Escherichia coli, Proteus vulgaris and Bacillus subtilis in instance. The results show that:extracts of D.Turcz. dictamnine and fraxinellone have better antibacterial activity.
