| ObjectiveIn this study,the patients with phlegm criteria and healthy volunteers in the prediabetes group were selected as the research objects.The syndrome and miscellaneous characteristics of the patients with phlegm syndrome in the early stage of diabetes were analyzed.The difference of intestinal flora between the patients with phlegm syndrome and healthy volunteers in the early stage of diabetes was compared.The relationship between the intestinal flora and phlegm syndrome was analyzed,and the possible role of intestinal flora in the occurrence and development of phlegm syndrome in the prediabetes period was explored.Methods34 patients with phlegm syndrome in pre diabetes were included in the phlegm syndrome group and 37 healthy volunteers were included in the normal group.Four diagnostic information and fecal samples were collected.Genomic DNA was extracted from their feces.16 Sr DNA sequence specific primers were used for real-time fluorescent quantitative PCR analysis.The two groups were compared to analyze the characteristics of intestinal flora in patients with phlegm syndrome in pre diabetes.In addition,the main TCM syndrome elements of phlegm syndrome group were extracted,and the correlation between the main TCM syndrome elements and the distribution of intestinal flora was analyzed.Results1.Distribution of syndrome elements: the distribution of syndrome elements in the disease position of phlegm syndrome group is ranked according to the proportion.The top three are spleen(76.47%),liver(67.65%)and kidney(32.35%);According to the proportion,the top five syndrome elements were phlegm(100%),dampness(75.53%),Qi Stagnation(64.71%),blood stasis(55.88%)and yin deficiency(52.94%).2.There were significant differences in intestinal flora community structure between phlegm syndrome group and normal group(P<0.01);Compared with the normal group,the number of species in phylum,class,order,family,genus and species in the phlegm syndrome group decreased,and the difference was statistically significant(P<0.01).3.At the phylum level,the composition of intestinal flora in phlegm syndrome group and normal group is very similar.Bacteroidetes and Firmicutes are the dominant bacteria in both groups.At the class level,the difference of actinomycetes between the two groups was statistically significant(P< 0.01).At the order level,there was significant difference between Burkholderia and bifidobacteria between the two groups(P<0.05,P < 0.01).At the department level,the difference between the two groups was statistically significant(P< 0.01,P <0.05,P<0.05).At the genus level,the difference of parabacteroides between the two groups was statistically significant(P< 0.05).4.There was a positive correlation between spleen element of disease location syndrome and the abundance of Prevotella(r=0.376,P< 0.05);There was a positive correlation between the abundance of kidney and Bacteroides(r= 0.357,P< 0.05);Cardiac syndrome element was positively correlated with the abundance of Bacteroides(r=0.418,P<0.05),and significantly negatively correlated with the abundance of macromonas(r=-0.459,P<0.01).There was a negative correlation between pathogenic syndrome element phlegm and the abundance of Bacteroides(r=-0.419,P<0.05),a positive correlation with the abundance of Prevotella(r=0.392,P<0.05),and a positive correlation with the abundance of macromonas(r=0.386,P<0.05);There was a negative correlation between dampness syndrome element and the abundance of parabacteroides(r =-0.371,P < 0.05).Conclusions1.The occurrence of phlegm syndrome in early diabetes is closely related to the spleen,liver and kidney.The main syndrome factors are phlegm,dampness,qi stagnation,blood stasis and yin deficiency.2.The number,diversity and relative abundance of intestinal flora in patients with phlegm syndrome in the early stage of diabetes have changed.3.There is a correlation between intestinal flora and syndrome element score. |
