| Objective:To explore the CXCR5 expression in lung adenocarcinoma and clinical relevance and CXCR5(+),CD8(+)T infiltration in the lung adenocarcinoma Righteousness.Methods:1.Download from the TCGA database and GEO database of patients with lung adenocarcinoma(LUAD)gene expression data set,Using R-studio to download data to clean up(TCGA including 535 samples of tissue adjacent to carcinoma tissue samples,59,of which 57 matching group,GSE31210 including 204 carcinoma tissue samples;GSE68465 including 82 carcinoma tissue samples),Analysis CXCR5 in tissue adjacent to carcinoma tissues and the expression of situation and CXCR5 expression and correlation of tumor size,sex,age and so on,Using ROC curve evaluation CXCR5 expression quantity of lung adenocarcinoma diagnosis efficiency,Combination of three groups of patients with clinical data and the expression of CXCR5 COX regression model is established,survival analysis.COX regression analysis for single factor and multiple factors.Another ss GSEA algorithm was used to study the expression of CXCR5(TCGA group)and the correlation of tumor immunity infiltration.2.choose in October 2020 to December 2021 in the first affiliated hospital of nanchang university 59 patients hospitalized with thoracic surgery Clinical pathologic specimens,collection of data,record patients’ age,sex,tumor size,lymph node metastasis.Using flow cytometry instrument flow analysis was carried out on the selection of tissue samples CXCR5(+),CD8(+)T in carcinoma infiltration degree of the organization(With CXCR5(+),CD8(+)T ratio for the amount of calculation),Combined with the tumor size,lymph node metastasis,gender,age,comparative analysis.Results:1.The matching samples and paired samples LUAD CXCR5 expression was higher than the tumor tissue adjacent to carcinoma CXCR5 expression,Study statistically significant difference(P < 0.001).2.CXCR5 expression and stage,tumor size,lymph node metastasis,the correlation of sex,age had no statistical significance.3.ROC curve evaluation CXCR5 expression efficiency to the diagnosis of lung adenocarcinoma,the results showed(Cutoff value of 0.108):TPR=0.671,TNR=0.712,PPV=0.955,NPV=0.193,About an index=0.383,AUC=0.676.4.Single and multi-factors COX regression analysis,the results showed that T stage,N stage,TNM staging,CXCR5 is independent prognostic factor in patients with lung adenocarcinoma,CXCR5 express high level of risk factors for prognosis.5.CXCR5 expression and B cells,T cells,plasma cells,dendritic cells(p DC),CD8 T cells and helper T cells and other immune levels were positively correlated6.CXCR5(+),CD8(+)T in the cancerous tissue infiltration degree is higher than that in tissue adjacent to carcinoma infiltration degree difference was statistically significant(P<0.001).CXCR5(+),CD8(+)T infiltration degree of correlation with tumor size was statistically significant,The greater the tumor CXCR5(+),CD8(+)T infiltration degree was higher.Conclusion:1.CXCR5 expression and the clinical correlation between lung adenocarcinoma,Found CXCR5 in LUAD significantly higher than the amount of expression in the tissue adjacent to carcinoma and CXCR5 lower expression is associated with better OS.CXCR5 as LUAD independent prognostic factors for patients2.CXCR5 expression quantity affect tumor immune microenvironment(CXCR5expression quantity and Bcells,T cells,how extensive the TFH and Cytotoxiccells positive phase),But the influence of the specific mechanism needs further research.LUAD patients with carcinoma of CXCR5(+),CD8(+)T infiltration degree is higher than that of tissue adjacent to carcinoma CXCR5(+)CD8(+)T degree of infiltration.LUAD tumor patients with larger CXCR5(+),CD8(+)T the higher degree of infiltrating. |
