| Objective To investigate the frequencies and the role of CD4+CXCR5+T cells in theCD4+T cells from peripheral blood and salivary gland of patients with primarySjogren’s syndrome (pSS)Methods Frequencies of blood CD4+CXCR5+T cell,CD19+CD27-na ve B cellã€CD19+CD27+memory B cell and CD19+CD27highplasmacell were analyzed by flowcytometry of24pSS patients before and after treatment and20healthy controls.Differences between groups and influences of treatment were analyzed. Meanwhile, theexpression of CD4+CXCR5+T cell,CD19+CD27-na ve B cellã€CD19+CD27+memory Bcell and CD19+CD27highplasmacell on salivary gland with patients of pSS withouttherapy was analyzed. Independent sample T test, person correlation analysis were usedfor statistical analysis.Resultsâ‘ The frequency of peripheral blood of CD4+CXCR5+in CD4+T cells andCD19+CD27-naive B cells in CD19+.B cells were increased in patients with pSS withouttreatment compared with healthy controls(17.90±4.40%vs.14.45±3.54%, P=0.019;(72.28±12.94%vs64.65±10.21%, P=0.043),).The percentage of peripheral blood ofCD19+CD27+memory B cells and CD19+CD27highplasma cells in CD19+B cells werereduced in patients with pSS without treatment compared with healthy controls(26.71 ±12.72%vs33.73±12.24%, P=0.059ï¼›1.08±0.58%vs1.61±0.62%, P=0.008).â‘¡Thefrequency of peripheral blood of CD4+CXCR5+Tfh cells had no correlations with Thefrequency of peripheral blood of CD19+CD27+memory B cells and CD19+CD27highplasma cells.â‘¢No correlations was found between clinical index such as ESR andpercentage of peripheral blood of CD4+CXCR5+T cells in patients with pSS withouttreatment(P>0.05).â‘£The frequency of salivary CD4+CXCR5+Tfh cells havesignificant correlation with CD19+CD27+memory B cells and CD19+CD27highplasmacells. And the percentages of salivary CD19+CD27highplasma cells have positivecorrelation with serum ANA titers.⑤The percentages of peripheral blood ofCD4+CXCR5+T cells were decreased ((15.19±3.23)%)after being treated for4weekswith prednisone (≤10mg/d) compared with those before receiving treatment((18.07±4.25)%).Conclusions These results demonstrate that the abnormality of CD4+CXCR5+T cellsmay play an important role in the pathogenesis of pSS. |
