| Objective:To observe the expression situation of renal tissue NF-κB and MCP-1 in rats with CRF after colon dialysis combined with Yishen Decoction,which provides a theoretical basis for the prevention and treatment of CRF.Method:Of the 60 healthy male SD rats,15 were randomly selected for the normal group,and the remaining 45 were filled with 2.5% adenine suspension according to the Yokozawa method,and a CRF model was established for consecutive 28 days at a dose of200mg/kg.d.And the CRF model was randomly divided into the model group,colon dialysis group,Yishen Decoction group.All 4 groups were given a free diet.Among the groups,the two treatment groups were given dialysis solution for colon dialysis,and the Yishen Decoction was given Yishen Decoction to retain enema.The treatment time lasts 4weeks.During the period,rats were weighed and recorded every week.When the time is over,we need to collect the rat arterial blood and serum creatinine,blood urea nitrogen,LPS and TNF-α were biochemically tested respectively to each group.At the same time,the kidney was removed for treatment,including making pathological section,HE staining,immunohistochemistry.Finally,the morphology and positive expression of NF-κB and MCP-1 were observed under a light microscope.The specific light density value is calculated by image processing software.The various data in the experimental process is analyzed by using the SPSS 26.0.Results:(1)After modeling,compared with the normal group,the modeled group was lean,the body hair brightness and the activity decreased,and the blood creatinine increased,the kidneys of rats enlarged and the surface of the uneven white particles.Microscopically,the glomeruli are atrophied,the tubules are dilated,atrophied and necrotic,and the renal interstitial fibrosis is infiltrated by inflammatory cells.(2)After modeling,compared with the normal group,the weight growth of modeled rats were slower.After 4 weeks of treatment,compared with the model group,the weight growth of the Yishen Decoction group was significant(P<0.05).There was no statistical significance between the colonic dialysis group and the model group(P>0.05).(3)After 4 weeks of study,the levels of blood Scr,BUN,LPS and TNF-α in model group were significantly higher than those in normal group(P<0.05).And compared with the model group,the blood Scr,BUN,LPS and TNF-α levels of the two treatment groups were significantly decreased(P<0.05).Compared with the colonic dialysis group,the blood Scr,BUN,LPS and TNF-α levels of Yishen Decoction group were decreased significantly(P<0.05).(4)Compared with the normal group,the model group showed atrophy of glomerulus,dilation,atrophy and necrosis of renal tubule,interstitial fibrosis of kidney and infiltration of inflammatory cells.Compared with the model group,the pathological injury of kidney in the two treatment groups decreased in different degree,especially in Yishen Decoction group.(5)Compared with the normal group,the expression of NF-κB and MCP-1 in the model group was increased(AOD P<0.05).The expression of NF-κB and MCP-1in two treatment groups were lower than those in model group(AOD P<0.05).The expression of Yishen Decoction group was lower than that in colon dialysis group(AOD P<0.05).Conclusion:Yishen Decoction may inhibit renal inflammation and fibrosis by down-regulating the expression of NF-κB and MCP-1 in renal tissue of CRF rats. |
