| Objectives:1.Through the study of literature,the Traditional Chinese Medicine connotation of intestinal-kidney axis is discussed,the intestine-kidney axis theory in Traditional Chinese Medicine is systematically analyzed and completed to expand the ideas of treatment and syndrome differentiation,and improve the guiding value for clinical treatment.2.The molecular mechanism of Taohechengqi Decoction in ameliorating the micro-inflammatory state of rats with chronic renal failure is further studied with the intestine-kidney axis as the new perspective and the TLRs/NF-κB classical inflammatory signaling pathway as the entry point,so as to consolidate the scientific basis of this prescription in the treatment of chronic renal failure.Methods:1.The theoretical study is mainly based on the literature review method: the literature is collected from the library of Fujian University of Traditional Chinese Medicine,and the literature and interpretation of related concepts related to intestine and kidney in classical works in various periods are collated through catalogue indexing and keyword searching,and analyzed through the chinese medicine theory to summarize the development,the physiological rules and pathological manifestations of the intestine-kidney axis theory in Traditional Chinese Medicine.The databases including CNKI and Wanfang are used as the platforms to supplement the modern research.2.The experimental study is conducted mainly based on the research method of animal experiments: 5/6 nephrectomized rats with chronic renal failure are used as the research vehicle,and 70 Wistar rats are used to collect relevant samples after modeling,grouping,and intervention administration.ELISA is used to detect serum Scr,BUN,IL-6,IL-10,TNF-α,IS,PCS,LPS,TMAO,and SIg A in feces.Western Blot is used to detect the protein expressions of claudin-1,ocludin,ZO-1,TLR2,TLR4,My D88,and NF-κB in intestine tissue.The m RNA of TLR2,TLR4,My D88,and NF-κB in intestine tissue is detected through real-time fluorescence quantitative PCR.The kidney and intestine histopathology is observed through light microscope,and the intestine histopathology is observed through transmission electron microscopy.Results:Theoretical research results:1.Through the Qin and Han dynasties,Jin and Tang dynasties,Song and Jin and Yuan dynasties,Ming and Qing dynasties,and late Qing dynasties to the Republic of China,based on the development and innovation of Chinese medicine theories in different contexts,the intestine-kidney axis theory in Traditional Chinese Medicine has been supplemented and deepened from the anatomical structure,physiology,pathology and treatment of related diseases of intestine and kidney.2.Physiological law of the intestine-kidney axis in Traditional Chinese Medicine: the intestine-kidney axis theory in Traditional Chinese Medicine can play a regulatory role in aqueous metabolism,the transformation of essence and blood,the opening and closing of urine and stool,and the lower-jiao qi-chenistry through the mutual cooperation between the intermediary complex associations.3.Pathological manifestation of the intestinal-kidney axis in Traditional Chinese Medicine: disorders of aqueous metabolism can be seen as edema,urinary inconvenience,thirst,phlegm,turbidity,etc;abnormal transformation of essence and blood can be seen as vomiting,abdominal distention,blood stasis,blood in urine,cloudy urine,blood in stool,deficiency,etc;disorders of the conduction of urine and stool can be seen as enuresis,nocturia,dysuria,diarrhea,constipation,etc.4.The intestine-kidney axis theory in Traditional Chinese Medicine refers to the two-way regulation between intestine and kidney under the guidance of Traditional Chinese Medicine theory,in which essence and fluid are the material basis of this function,and qi-chemistry is the driving force,and its academic connotation can be summarized as“absorbing and changing essence and fluid,qi-chemistry to use”.Experimental study results:1.Experiment 1: Effect of Taohechengqi Decoction on the general status and micro-inflammatory status of rats with chronic renal failure:Taohechengqi Decoction can reduce serum SCR and BUN levels,lower serum IL-6 and TNF-α levels and increase serum IL-10 expression in rats with chronic renal failure,and reduce renal pathological changes such as glomerular edema,thylakoid matrix accumulation and renal tubular dilatation.2.Experiment 2: Effect of Taohechengqi Decoction on the intestinal barrier of rats with chronic renal failure:Taohechengqi Decoction reduce the levels of serum IS,PCS,LPS and TMAO in rats with chronic renal failure,stimulate SIg A,delay the damage of claudin-1,occludin and ZO-1 in intestinal tissues,and maintain the epithelial cells and tight junction structure of intestinal tissues.3.Experiment 3: Regulation of TLRs/NF-κB signaling pathway in intestinal tissues of rats with chronic renal failure by Taohechengqi Decoction:Taohechengqi Decoction reduced the m RNA and protein expression of TLR2,TLR4,My D88,and NF-κB in the intestinal tissues of rats with chronic renal failure.Conclusions:1.The intestine-kidney axis theory in Traditional Chinese Medicine,which has been constructed from the occurrence and development of the theory,the physiological rules and pathological manifestations of intestine and kidney,and modern research,reflects the mechanism of two-way regulation between intestine and kidney in a positive and negative direction.Its connotation in chinese medicine can be summarized as “absorbing and changing essence and fluid,qi-chemistry to use”.2.Based on the intestine-kidney axis theory,the molecular mechanism of Taohechengqi Decoction to ameliorate the micro-inflammatory state in rats with chronic renal failure may be related to the ability of Taohechengqi Decoction to reduce the expression of pro-inflammatory factors and endotoxin,improve the intestinal barrier function,and inhibit the intestinal TLRs/NF-κB signaling pathway.Innovation Points:1.Theoretical innovation: based on the western medical concept hypothesis intestinal-kidney axis,the intestine-kidney axis theory in Traditional Chinese Medicine and the academic connotation of “absorbing and changing essence and fluid,qi-chemistry to use”are summarized,which is a new exploration to promote the modern research and innovation of Traditional Chinese Medicine theory.2.Experimental innovation: to investigate the mechanism of the treatment of rats with chronic renal failure with the classic prescription Taohechengqi Decoction from both intestinal and renal perspectives,and focusing on the effect of the intestinal barrier and the regulation of intestinal TLRs/NF-κB pathway,and to provid new ideas and basis for the identification and treatment of difficult diseases with the classic prescription. |
