| Objective: To investigate the expressions of proangiogenic factor VEGF and S100A2 in triple negative breast cancer(Triple Negative Breast Cancer,TNBC),and analyze their clinical correlation with TNBC.Methods:(1)The general clinical data of 51 patients with TNBC diagnosed by pathology department of our hospital were collected,the expression of VEGF in TNBC tissue was detected by immunohistochemistry and its correlation with clinical was analyzed.(2)Download the required raw data information of TNBC and normal breast tissue from the GEO database,use R language and related data analysis packages to perform single-cell RNA sequencing of the data,and analyze the differential expression of VEGF and S100A2 in TNBC and normal breast tissue.(3)30 fresh cancer tissues and 30 paracancerous tissue specimens from Affiliated Hospital of Youjiang Medical College for Nationalities who underwent surgical treatment and were pathologically diagnosed as TNBC were collected.HE staining and Masson staining were used to understand the structural differences between TNBC and paracancerous tissues.Immunofluorescence staining was used.The expression of vascular endothelial factor VEGF and S100A2 in TNBC and adjacent tissues were detected by method,and their clinical correlation was analyzed.Results:(1)Immunohistochemical results of 51 TNBC patients showed that VEGF expression was significantly correlated with lymph node metastasis and distant metastasis(P<0.05);There was no significant difference in age,tumor size and histological grade(P>0.05).(2)The results of single-cell RNA sequencing showed that the m RNA expressions of VEGF and S100A2 in TNBC were significantly higher than those in normal breast tissue.The results of subpopulation analysis showed that VEGF was mainly expressed in endothelial cells,while S100A2 was mainly expressed in epithelial cells and a small amount in endothelial cells.(3)The results of HE and Masson staining showed that there were a large number of hyperchromatic nuclei in TNBC,which were piled into clusters,the internal structure was disordered,and the normal breast structure disappeared.Rarely,nuclei accumulate in clusters.(4)Immunofluorescence results showed that the expressions of VEGF and S100A2 in TNBC tissues were significantly higher than those in adjacent tissues,and the difference was statistically significant(P<0.05).Expressions of VEGF and S100A2 in TNBC are related to lymph node metastasis.The expression of S100A2 in TNBC tissues was significantly correlated with tumor diameter,but not with age and histological grade.There was no correlation between the expression of VEGF and age,tumor diameter and histological grade(P>0.05).Conclusion:(1)Analysis of clinical data found that the expression of VEGF in TNBC was associated with lymph node metastasis and distant metastasis.(2)The expression levels of VEGF and S100A2 in TNBC were significantly higher than those in adjacent tissues in GEO dataset.(3)Experimental sample verification found that the expression of VEGF and S100A2 in TNBC was related to lymph node metastasis,S100A2 was related to tumor size.(4)High expression of VEGF and S100A2 are risk factors for overall survival of TNBC and can be used as prognostic indicators. |
