| Objective: To select differential genes in triple negative breast cancer and analyze the effect of differential genes on the proliferation of triple negative breast cancer cells,and to study its molecular mechanism.Methods: Gene expression omnibus(GEO)database was used to analyze the gene expression.The GEO database of the National center for biotechnology information(NCBI)was selected as Triple Negative Breast Cancer,The Fold change and P value of the original data downloaded from GEO database were calculated using DEseq in R software.The enrichment analysis of GO and Kyoto Encyclopedia of Genes and Genomes(KEGG)was performed using DAVID database.Bioinformation analysis: The expression of Rab27 A and ZEB1 in breast cancer was analyzed using the Tumor Genome Atlas(TCGA)database.Through literature review,it was found that IL-6/STAT3 signaling pathway plays an important role in regulating tumor epithelial-mesenchymal transition and affecting immune microenvironment.Western Blot experiment verified the regulatory relationship between Rab27 A,ZEB1 and IL-6/STAT3 signaling pathways.Results: GEO database was used to screen Rab27 A gene as a potential research object.The differences between Rab27 A and ZEB1 between normal people and patients were analyzed according to TCGA data.The differences between Rab27 A and ZEB1 in other malignant tumors were statistically significant.The experiment verifies that there is an upstream and downstream regulation relationship among the three.Conclusion: Rab27 A can activate IL-6/STAT3 signaling pathway to promote the proliferation of triple negative breast cancer through positive regulation of ZEB1 expression. |
