| Objective:This study attempts to construct the spinal cord injury model of fully transected rats,explore the changes and laws of Rac1 protein expression in spinal cord injury rats after Jiaji electroacupuncture intervention,observe the histopathological changes in the injured area and the improvement of motor function before and after treatment,and then explore the mechanism of Jiaji electroacupuncture in the treatment of spinal cord injury in rats,so as to provide a reliable experimental basis for clinical treatment.Method:Firstly,81 adult female SD rats were selected.According to the method of random sampling digital table,the rats were divided into different groups:sham operation group,model group and Jiaji electroacupuncture group.The number of rats in each group was equal.Secondly,the rat model of total transverse spinal cord injury was made.The sham operation group needed to implement anesthesia on the rats,and then cut their back skin and muscle tissue and occlude the lamina to expose the T10 spinal cord,not give transection.After that,the cut wound was sutured,and only routine nursing was given after operation;The spinal cord was transected at the T10 horizontal segment in both spinal clip electroacupuncture group and model group,After the successful preparation of the model,the model group did not receive treatment,but only received routine postoperative care;The Jiaji electroacupuncture group needed to carry out Jiaji electroacupuncture treatment for 30 minutes,and then once a day for 30 minutes.The rats in the sham operation group and the model group grabbed once a day.The motor function of the rats in each group was evaluated by BBB score on 3d,7d and14 d after operation,Then,the spinal cord injury tissues in the spinal cord injury area of rats in each group were taken out respectively.The expression of Rac1 protein in each group was measured by Western blot,and the pathological changes of spinal cord were explored by the application of HE staining.Results:1.BBB score: the BBB score of each time point in the sham operation group was 21;In contrast,the BBB scores of the other two groups decreased at three time points(P < 0.05),and the decline was the most obvious in the model group;Compared with the model group,the BBB scores of Jiaji electroacupuncture group increased at three time points after treatment,and the difference had certain statistical significance(P< 0.05);The comparison within the group showed that the BBB scores increased at 7 and 14 days of treatment compared with 3 days of treatment,and the difference had statistical significance(P< 0.05).2.Results of Western blot: At the time points of 3D,7d and 14 d after operation,the lowest expression of Rac1 protein in the three groups was the sham operation group,and the expression of Rac1 protein in the other two groups was relatively high.After treatment,the expression of Rac1 protein in Jiaji electroacupuncture group increased to a certain extent.Compared with the model group and the sham operation group,the expression of Rac1 protein in the former group was significantly increased,and the difference was statistically significant(P < 0.05);Compared with the model group,the increase of Rac1 protein expression was reflected in all time points of Jiaji electroacupuncture group,and the difference was statistically significant(P< 0.05);Comparison of Jiaji electroacupuncture group;Compared with the 3d group of Jiaji electroacupuncture group,the 7d group and 14 d group were significantly improved,and the difference was statistically significant(P <0.05).3.HE staining results:among the three groups,the sham operation group had the highest integrity and clarity of tissue structure.The central canal of rat spinal cord showed normal structure,with neat arrangement of gray and white matter,clear boundary range,balanced distribution of neurons and glial cells,round cell body,normal morphology of glial cells,no necrosis and edema.Model group in 3 day,the structure of spinal cord tissue was incomplete,the destruction was obvious,the number of neurons swelled and necrotic increased,and inflammatory cells could be seen in the spinal cord tissue;On the 7th day,the spinal cord tissue was seriously damaged,the residual cell body swelled and deformed,the number of glial cells and inflammatory cells increased,and the loss of neurons was obvious;On the14 th day,there was focal liquefied necrosis and the formation of nerve cell vacuoles.Only a small amount of residual neuronal tissue could be seen at the distal end of the injured tissue.Most neurons had shrunk and disappeared,inflammatory cell infiltration and glial cell proliferation.At 3d,7d and 14 d in Jiaji electroacupuncture group,with the increase of treatment time,the number of neurons increased,the phenomenon of interstitial edema improved significantly,the situation of structural looseness and tissue loss improved,the inflammatory cells decreased gradually,and the degree of infiltration and damage of inflammatory lesions decreased step by step.Conclusion:1.Jiaji electroacupuncture can promote axon regeneration and improve the nerve and motor function of rats.The mechanism may be related to the up-regulation of RAC1 protein expression in injured area.2.Jiaji electroacupuncture can play a neuroprotective role and promote the repair of injured spinal cord in rats. |
