Objective:In this study,the spinal cord of rats was completely transected,and after the intervention of Jiaji electroacupuncture,the motor function of rats with spinal cord injury was observed,and the neuronal morphology changes of spinal cord injury and the expression of MIF protein in spinal cord injury rats were analyzed.Furthermore,it provides some experimental basis for the study of Jiaji electroacupuncture in the treatment of spinal cord injury.Method:In this experiment,a total of 100 SD adult female rats were prepared,and81 rats were used in the specific experiment.Sham operation group(experimental control group),model group and Jiaji electroacupuncture group had 27 rats in each group.There were 9 rats in each experimental group at three time points of 3d,7d and 14 d.First of all,the rats were anesthetized for modeling.In the sham-operated group,the back skin of the rats was incised at T10 of the spine,and the subcutaneous tissue was separated.Then only the lamina was removed to expose the spinal cord,and the wound was sutured layer by layer without transection.The T10 segment of spinal cord of rats must be completely transected in both the model group and Jiaji electroacupuncture group,and the wound was sutured layer by layer.Secondly,after the modeling was completed,the Jiaji electroacupuncture group received Jiaji electroacupuncture treatment for 14 consecutive days,once a day for 30 minutes each time.While the sham operation group and the model group only performed the experimental movements,and did not receive spinal electroacupuncture treatment.The BBB scores of each group before and after modeling at 3,7 and 14 days were recorded during the experiment to assess the motor function of both lower limbs of the rats.Then the spinal cord injury tissues of rats in each group were intercepted at three time points,and the neuronal morphology changes of spinal cord injury tissues were analyzed by Nissl staining and the expression of MIF protein was analyzed by Western Blot method.Results:1.BBB score: The 3d gait of the sham operation group basically recovered after modeling,and the motor function was completely the same as before modeling.Therefore,the BBB score was 21 points at different time points,which was not statistically significant(P>0.05).The BBB scores of the model group and Jiaji electroacupuncture group were lower than those of the sham operation group on the 3,7 and 14days(P<0.05).However,the BBB scores of the Jiaji electroacupuncture group were higher than those of the model group at three time points(P<0.05),and the BBB scores of the Jiaji electroacupuncture group were significantly different from those of the model group at 14 days of treatment.The model group and the Jiaji electroacupuncture group were compared within groups,and the BBB scores at different time points were significantly different(P<0.05).2.Nissl staining results: at the three time points of 3,7,and 14 days after modeling,the neurons in the tissue sections of the sham operati on group were blue,with clear structures,and large and round cell bodies.The Nissl bodies in the cell body are blue-purple,numerous and evenly distributed.There was no significant change in the average optical density(AOD)at three time points in the sham group(P>0.05).Compared with the sham operation group,the neuron staining of the tissue sections of the model group became lighter,the cells were vacuolated,edema and necrosis,and the Nissl bodies in the cell body were lighter,stained unevenly,decreased in number,or even disappeared,and 3days,The AOD values at the three time points of 7 and 14 days all decreased significantly(P<0.05).In the model group,as time went on,the neuron damage became more serious,the number of Nissl bodies dec reased,and the AOD value also decreased(P<0.05).Compared with the model group,after electroacupuncture treatment in the Jiaji electroacupuncture group,the neurons in the tissue sections stained darker,the cell body gradually became larger and fuller,and the Nissl bodies in the cell body stained deeper and increased in number.The AOD values at the three time points all increased significantly(P<0.05),but were still significantly lower than the level of the sham operation group(P<0.05).With the increase of the three treatment time points of 3,7 and 14 days,the morphology of neurons in the Jiaji electroacupuncture group gradually tended to normal,and the AOD value increased significantly(P<0.05).3.Western Blot detection results: the expression level of MIF protein at each time point of 3,7 and 14 days in the sham operation group was the lowest,and the expression level of MIF protein at the three time points had no significant change(P>0.05).The expression levels of MIF protein in the model group and the Jiaji electroacupuncture group at three time points of 3,7 and14 days were significantly higher than those in the sham operation group,and the difference was statistically significant(P<0.05).After treatment at three time points,the expression level of MIF protein in the Jiaji electroacupuncture group was significantly lower than that in the model group,and the difference was statistically significant(P<0.05).In the model group,with the prolongation of the spinal cord injury time,the expression levels of MIF protein at the three time points increased step by step,and the difference was statistically significant(P<0.05).The expression levels of MIF protein in the Jiaji electroacupuncture group gradually decreased at three time point s with the prolongation of treatment time,and the difference was statistically significant(P<0.05).Conclusion:1.Jiaji electroacupuncture can improve the motor function of rats with total transected spinal cord injury,and promote the regeneration of n eurons in the rat spinal cord injury area.2.Jiaji electroacupuncture can reduce the expression of MIF protein in the rat spinal cord injury area,indicating that it may be related to the mechanism of Jiaji electroacupuncture treatment of rat spinal cord injury. |
