Study On The Material Basis And Quality Standard Of Paeoniae Radix Alba

Paeoniae Radix Alba is the dried root of Paeonia lactiflora,which was first recorded in the Shennong′s Classic of Materia Medica.It has the effects of nourishing blood and regulating menstruation,softening liver and relieving pain,calming liver Yang,astringeting Yin and stopping sweating.It is commonly used to treat rheumatoid arthritis,visceral pain,chronic gastritis,chronic hepatitis B and dysmenorrhea.The current research of Paeoniae Radix Alba focuses on the improvement of quality standards,clinical medication and pharmacodynamics,but the previous pharmacodynamics research is not associated with the theory of traditional Chinese medicine,and limited by the traditional research tools,research ideas and research techniques,the theoretical research on the properties of five flavors of traditional Chinese medicine is not perfect.Therefore,under the theoretical system of traditional Chinese medicine,based on the core concept of quality marker,from the perspective of"effectiveness"and"Property-Efficacy-Material basis"ternary theory,this thesis takes Paeoniae Radix Alba as the research object to explain the material basis of Paeoniae Radix Alba and related pharmacological mechanisms at the molecular level.To provide the basis for the quality evaluation,disease diagnosis and clinical practice of Paeoniae Radix Alba a,and also to provide the reference for the further study of traditional Chinese medicine theory,This thesis conduct the following researches:1.In this study,HPLC-Q-TOF-MS/MS technology was used to identify and characterize a total of chemical components in Paeoniae Radix Alba,roasted Paeoniae Radix Alba and Paeoniae Radix Rubra.42 compounds were identified and characterized from Radix Paeoniae Alba,including 26 monoterpenes and their glycosides,1 flavonoid,7 tannins,6 organic acids and 2 other compounds.43 compounds were identified and characterized from roasted Paeoniae Radix Alba,including 27 monoterpenes and their glycosides,1 flavonoid,7 tannins,6 organic acids and 2 other compounds.35 compounds were identified and characterized from Paeoniae Radix Rubra,including 21 monoterpenes and their glycosides,1 flavonoid,6 tannins,6 organic acids and 1 other compounds,and the material basis of Paeoniae Radix Alba,roasted Paeoniae Radix Alba and Paeoniae Radix Rubra was clarified.2.The theoretical system of traditional Chinese medicine holds that"medicinal property"is also the core part of the traditional efficacy of traditional Chinese medicine.therefore,based on the scientific connotation of"taste"in the properties of traditional Chinese medicine,the molecular docking technology is adopted in this chapter to establish the ligands library of bitter and sour docking ligands of Paeoniae Radix Alba,homology modeling was carried out by selecting positive ligand of each flavor,and the material basis of medicinal property of Paeoniae Radix Alba was determined from the perspective of real"taste".The results showed that there were 13 chemical components in the root of Paeoniae Radix Alba among which monoterpenes and their glycosides were the main components.There were 14 chemical components in the root of Paeoniae Radix Alba among which tannins were the main component.3.Based on the previous molecular docking results and related literature,17 compounds including the main components of Paeoniae Radix Alba the components entering the blood and the main structural types were selected as the research objects,with the help of TCMSP,Swiss Target Predicition and Uni Prot database,and then the pathway enrichment analysis and bioinformatics analysis were carried out by using STRING 10 database and Omicsbean online analysis platform.Finally,the"compound-target-pathway-pharmacological action-efficacy"network diagram was constructed by Cytoscape software,and the material basis of property/efficacy of Paeonia lactiflora was predicted and analyzed from the perspective of biological effect to preliminarily explain the pharmacological mechanism of its traditional efficacy.4.Based on the prediction results of network pharmacology,combined with relevant literature reports,we established an in vitro inflammatory model of RAW264.7 cells induced by LPS,At the same time,estrogen receptor（ERα）and muscarinic receptor（M2）related to nourishing blood and regulating menstruation;cyclooxygenase-2（COX-2）,nuclear factor kappa B（NF-κB）,Toll-like receptor（TLR4）,prostaglandin E₂（PGE₂）related to softening liver-yang and relieving pain were selected;endothelin receptor（ET_B）related to calming liver-yang.Adrenergic receptors（α1A）and muscarinic receptors（M3）related to antiperspiration were used as research carriers.The binding experiments of chemical constituents and receptors were carried out to screen the active chemical components.From the point of view of efficacy,it is clear that paeoniflorin,Albiflorin,oxidized paeoniflorin,Gallic acid,catechin,pentagalloyl glucose,Gallic acid and benzoyl paeoniflorin may be the efficacy material basis of Paeoniae Radix Alba.It can be identified as a quality marker of Paeoniae Radix Alba.5.Based on the results of the previous four chapters,the fingerprint was established to evaluate the quality consistency of Paeoniae Radix Alba,and the"one test and multiple evaluation"content determination method was established to improve the quality standard of Paeoniae Radix Alba.To sum up,42 chemical constituents were identified and characterized from Paeoniae Radix Alba,On this basis,based on the core concept of quality markers,that is,from the point of view of the complete expression of"effectiveness",this thesis carries on the validity correlation from the aspect of medicinal properties,and takes the ternary theory of"Property-Efficacy-Material basis"as the research thought.the material basis of medicinal properties of Paeoniae Radix Alba was clarified from the point of view of real"taste"and"efficacy five flavors",the relevant pharmacological action mechanism was preliminarily explained,and the quality markers of Paeoniae Radix Alba were determined.Finally,the quality evaluation method of Paeoniae Radix Alba was established by fingerprint and multi-evaluation technology,which provided a scientific basis for the follow-up improvement of the quality standard of Paeoniae Radix Alba.
