| Herb couple is an important component of traditional Chinese medicine prescriptions, which is different from the single herb medicine, and it has a fixed combination rather than the complex of prescriptions. The drugs play a role in organism through a multi-channel, multi-target and integrated regulation work, and its composition is complex, which makes the study of group-effect of the drug compatibility needs for entirety and syst. So, it is required to introduce new research tools to deal with complex systems.In this paper, the study object is the material basis and group-effect of the herb couple of Radix Bupleuri and Radix Paeoniae Alba, using the ideas and methods of traditional Chinese medicine chemics to find the active ingredient and mechanism. The main study includes the following aspects:(1) Standardized component groups of Radix Bupleuri and Radix Paeoniae Alba were prepared using the modern technologies of extraction, separation and preparation of active ingredients in traditional Chinese medicine. And, the main substances in the component groups were identified using high performance liquid chromatography -mass spectrometry (HPLC-MS) and other modern analytical techniques.(2) The components with the activity of liver cell protection or influence of cell proliferation were screening using rat liver cell injury model and mouse spleen cell proliferation model. By combining the screening results with the analysis data of components, the active ingredients in Radix Bupleuri were determined to be mainly flavonoids and saponins, while the active ingredients in Radix Paeoniae Alba are mainly monoterpene saponins.(3) The effects of different components of Radix Bupleuri and Radix Paeoniae Alba herb couple on hepatoprotection and immunization were studied using acute liver injury mice model and ear swelling mice model. It was found that the compatibility of Bupleurum flavonoids and total glucosides of paeony has a similar effect with the extraction of Radix Bupleuri and Radix Paeoniae Alba.(4) A high performance liquid chromatography (HPLC) method for the simultaneous determination of albiflorin, paeoniflorin, naringin and glycyrrhizin in Qizhiweitong granules(contains Radix Bupleuri, Radix Paeoniae Alba and other four herbs) was established. |
