Objective: To analyze the correlation between miRNA-505 and prostate cancer,explore its possible mechanism of action in prostate cancer.Methods:(1)To analyze the expression of miRNA-505 in prostate cancer tissues and its correlation with prostate cancer stage and grade,based on the miRNA expression profile and clinical data of prostate cancer patients in TCGA database.(2)Collected the clinical data,blood samples and biopsy tissue samples of patients undergoing prostatic puncture in the Department of Urology,the First Affiliated Hospital of Xinjiang Medical University from March 2021 to June 2021,detected the expression levels of miRNA-505 in blood and tissues of patients,to analyze the expression differences and diagnostic value of miRNA-505 in prostate cancer combined with clinical data.(3)Up-regulated the expression of miRNA-505 in prostate cancer cell,the detected the changes of physiological function,glycolysis level and expression of HMGB 1,PTEN and glycolysis related genes.Results:(1)Expression data of miRNA-505 in 551 prostate cancer and non-cancer tissues from TCGA database showed that miRNA-505 is down-regulated in prostate cancer tissues(P<0.05),according to the data of 278 cases of prostate cancer in the database,miRNA-505 expression in tissues is lower in prostate cancer patients with higher malignant degree of tumor(P<0.05).(2)We collected 26 clinical tissue samples in this study,including 13 tumor tissues and 13 benign hyperplasia tissues.T-test of two independent samples showed that the expression level of miRNA-505 in tissues was lower than that in benign prostatic hyperplasia patients(P<0.05),collected a total of 93 patients’ blood samples,including 41 patients with prostate cancer and 52 patients with benign prostatic hyperplasia.Multivariate logistic analysis showed the expression level of miRNA-505 in blood >0.68 is a protective factor for prostate cancer(P<0.05),and there is a linear relationship between miRNA-505 in blood and the risk of PCa(overall association P<0.01,nonlinear P>0.05).(3)The expression level of miRNA-505 in prostate cancer cells is lower than that in normal prostate cells.Upregulation of miRNA-505 expression in prostate cancer cells showed that cell glycolysis level is decreased,and the proliferation,migration and invasion ability of tumor cells are decreased,apoptosis level is increased.Meanwhile,HMGB 1 gene expression is down-regulated and PTEN gene expression is up-regulated.The expression of glycolysis related gene PGK 1 is decreased.Conclusion: miRNA-505 is down regulated in the blood and tissues of prostate cancer patients,and its expression level is correlated with the degree of malignancy of prostate cancer.miRNA-505 can down-regulate the expression of HMGB 1 and PGK 1 in prostate cancer cells,up-regulate the expression of PTEN and inhibit the glycolysis level of tumor cells,ultimately inhibit the proliferation,migration,invasion and ability of tumor cells,and promote cell apoptosis.
