| Purpose: The significance of the different differences and changes of spleen diseases was discussed by observing the spleen index,peripheral blood and spleen T lymphocyte subset changes,apoptosis of splenocyte and spleen pathological changes in the low dose irradiation group,repeated low dose irradiation group,high dose irradiation group and blank control group after radiation irradiation of the spleen region.Methods: After irradiating the spleen region of mice with different doses of radiation,mouse models of each experimental group were established.The mice were sacrificed humanely,and peripheral blood and spleen tissues were collected.The changes of spleen index were calculated,the changes of peripheral blood and spleen T lymphocyte subsets(CD3+T lymphocyte,CD4+T lymphocyte,CD8+T lymphocyte)and apoptosis of spleen cells were analyzed by flow cytometry,and the pathological changes of spleen tissues were analyzed by HE staining.Results: Compared with blank control group,low dose irradiation group and high dose irradiation group,spleen index of mice in low-dose irradiation group was significantly increased;Low dose irradiation,repeated low dose irradiation group mice peripheral blood and spleen of CD3 + T lymphocytes,CD4 + T lymphocytes percentage in the blank control group and high dose irradiation group significantly increased,low dose irradiation group,low dose irradiation group,high dose irradiation group of peripheral blood and spleen CD8 + T lymphocytes in mice were significantly lower proportion of the compared with blank control group,The apoptosis rate of spleen cells in high dose irradiation group was significantly higher than that in blank control group,low dose irradiation group and repeated low dose irradiation group.Different exposure group of mice spleen HE pathological dyeing are changed compared with blank control group,low dose irradiation and repeated low dose irradiation group mainly for splenic white pulp hyperplasia,bigger volume,number of lymphocytes is rich,and high dose irradiation group mainly for lymphocyte is loose,visible lymphocyte apoptosis,cell nucleus pycnosis,disappear.Conclusion: By low dose,after repeated low dose irradiation in mice spleen area,peripheral blood and spleen T lymphocyte subsets changes,makes the CD3 + T lymphocytes,CD4 + T cells express higher proportion,low doses of spleen area light to stimulate the spleen immune reaction,and after high dose irradiation in mice spleen,CD3+ T lymphocytes,CD4 + T lymphocytes decreased significantly,Stimulate the apoptosis of spleen and inhibit the occurrence of spleen immune response. |
