ObjectiveAllergic fungal sinusitis is a subspecies of chronic rhinosinusitis. In recent years, the research on allergic fungal sinusitis tends to be the etiology theory of multi-factor interaction, and inflammatory cytokines play a role in this process. This experiment aims to study the expression of cytokine interleukin in the serum of patients with allergic fungal sinusitis,to explore its possible pathogenesis of allergic fungal sinusitis, and to provide new ideas for clinical treatment of allergic fungal sinusitis.MethodsExperimental group: 26 patients with allergic fungal sinusitis who received surgical treatment in the Department of Otorhinolaryngology head and Neck Surgery,Affiliated Hospital of Qingdao University from September 2019 to February 2022 and met the diagnostic criteria were selected.Control group: 30 patients admitted to the department of Otolaryngology,Head and neck Surgery,Affiliated Hospital of Qingdao University for nasal bone fracture and underwent closed reduction for nasal bone fracture during the same period were collected. Methods: Fasting blood was collected from patients, and serum levels of interleukin-2,interleukin-4,interleukin-5,interleukin-6,interleukin-8,interleukin-10 and interleukin-17 were detected by ELISA,and correlation analysis was conducted. The severity of the patients in the experimental group was assessed by Lund-Mackay method, and the correlation between the levels of interleukins and CT scores of the nasal sinuses was analyzed.χ2 test was used for counting data,Mann-Whitney U test was used for measurement data, and Spearman rank correlation analysis was used for correlation analysis.P < 0.05 was considered statistically significant.Results1.The serum IL-2 concentration in the control group was 2.09(1.33-3.73)pg/ml,and that in the experimental group was 1.77(1.08-2.33)pg/ m L.The expression of IL-2 in the experimental group was lower than that in the control group, and the difference was significant(P=0.041<0.05).CT score of sinus in the experimental group was 9.5(5-14),and there was a significant negative correlation between CT score of sinus and IL-2concentration in the experimental group(r =-0.488,P=0.011<0.05).2.The serum IL-4 concentration in the control group was 1.28(0.74-1.44)pg/ m L,and that in the experimental group was 1.96(1.29-3.73)pg/ml.The serum IL-5concentration in the control group was 1.10(1.10-1.69)pg/ m L,and that in the experimental group was 1.94(1.50-3.75)pg/ml.The serum IL-17 concentration was 1.52(0.95~2.14)pg/ m L in the control group and 2.14(2.00~2.40)pg/ml in the experimental group. The expressions of IL-4,IL-5 and IL-17 in the experimental group were higher than those in the control group, and the differences were statistically significant(P values were 0.001,0.000 and 0.001,respectively,all <0.05).Sinus CT score was positively correlated with the concentration of IL-4,IL-5 and IL-17 in the experimental group, and the correlation was significant(r value 0.568,0.413,0.516,P value 0.002,0.036,0.007,all <0.05).3.The serum IL-6 concentration in the control group was 1.90(0.72-2.75)pg/ m L,and that in the experimental group was 1.93(1.39-2.56)pg/ml.The serum IL-8concentration was 2.30(1.39-2.30)pg/ m L in the control group and 2.30(1.40-3.47)pg/ml in the experimental group. The serum IL-10 concentration in the control group was 1.08(0.92-1.09)pg/ml,and that in the experimental group was 0.94(0.71-1.09)pg/ml.The expression of IL-6 and IL-8 in the experimental group was higher than that in the control group, while the expression of IL-10 was lower than that in the control group. However, there was no significant difference(P = 0.657,0.397,0.183,all >0.05).The CT score of sinus in the experimental group was 9.5(5-14),and there was no significant correlation between the CT score of sinus and the concentration of IL-6,IL-8 and IL-10 in the experimental group(P values were 0.873,0.353 and 0.578,respectively,>0.05).Conclusion1.The expression level of IL-2 in the experimental group was significantly lower than that in the control group. There was a significant negative correlation between the expression level of IL-2 in the experimental group and the sinus CT score of patients, suggesting that IL-2 plays an inhibitory role in the occurrence and development of allergic fungal sinusitis, and the content of IL-2 was negatively correlated with the severity of the disease in the experimental group.2.The expression levels of IL-4,IL-5 and IL-17 in the experimental group were significantly higher than those in the control group, and there was a significant positive correlation between the expression levels of IL-4,IL-5 and IL-17 in the experimental group and the sinus CT scores of patients, suggesting that they play a promoting role in the occurrence and development of allergic fungal sinusitis. The levels of IL-4,IL-5 and IL-17 were positively correlated with the severity of the disease in the experimental group.3.The expressions of IL-6,IL-8 and IL-10 in the serum of the control group and the experimental group were different, and the differences were not statistically significant, suggesting that the occurrence of allergic fungal sinusitis was related to IL-6,IL-8 and IL-10,and there was no significant correlation between the expression levels of IL-6,IL-8and IL-10 in the experimental group and the sinus CT score of patients.
