| Objective:To investigate the correlation between the long non-coding RNA H19 and H19 single nucleotide polymorphism(SNP)of imprinted genes and the occurrence of recurrent spontaneous abortion(RSA),and to provide a theoretical basis for a deeper understanding of the pathogenesis of recurrent abortion.The study was carried out to establish the theoretical basis for the screening,diagnosis,and treatment of recurrent abortion.Research methods:Chorionic and meconium tissues from 34 patients with recurrent spontaneous abortion and 42 patients with voluntary termination of pregnancy in normal early pregnancy from January 2020 to July 2021 in Weihai Maternal and Child Health Hospital affiliated with Qingdao University were selected for the study.The H19 SNP loci were searched using the database,and six SNP loci were included after screening: rs217727,rs2071095,rs2107425,rs3741219,rs2067051,and rs7103849.Genomic DNA was extracted from the study subjects,and the ltered SNPs were genotyped using gene sequencing.A genetic equilibrium test was performed using the Hardy-Weinberg law to determine whether the study subjects were representative of the sample.The differences in age,gestational week,and BMI between the two groups were compared by t-test and chi-square test.RT-PCR was used to detect the expression of LncRNA H19 in the chorionic and meconium tissues of the two groups,and then to determine the effect of each genotype of SNP on the expression level of H19.Results:1.The frequency of the H19 gene rs217727 GG genotype was 55.8% in the RSA group,which was higher than that of the control group by 26.2%,and the frequency of the G allele was 74.6%,which was higher than that of the control group by 50%.The differences in genotype and allele distribution at this locus in the two groups were statistically significant(p<0.05).rs2071095,rs2107425,rs3741219,rs2067051,rs7103849 locus genotype and allele distribution of H19 gene were not statistically significant(p>0.05).2.The expression of LncRNA H19 in the chorionic tissue of patients in the RSA group was significantly higher than that of LncRNA H19 in the control group(p<0.05);the expression of LncRNA H19 in the meconium tissue of patients in the RSA group was not significantly different from that of LncRNA H19 in the control group(p>0.05).3.The rs217727 locus GG and AG+GG genotypes could increase the expression level of chorionic H19 in RSA patients(p<0.05);rs2071095,rs2107425,rs3741219,rs2067051,rs7103849 genotypes were not significantly correlated with the expression level of chorionic H19 in RSA patients(p> 0.05).Conclusions:1.rs217727 locus SNP of the H19 gene was associated with the occurrence of recurrent miscarriage.Rs2107425,rs3741219,rs2071095,rs2067051,and rs7103849 loci of the H19 gene were not significantly correlated with the event of recurrent miscarriage.2.H19 gene expression was elevated in the chorionic tissue of patients with recurrent miscarriage,and the rs217727 GG,AG+GG genotype was associated with high expression of the chorionic H19 gene in RSA patients. |
