| Background Recurrent spontaneous abortion(RSA),which is a common pathological pregnancy disorder, has negative effects on the patients’ physical and mental health and their families. The etiology is so complex that 25% to 50% of patients do not find a cause.Existing studies have shown that single nucleotide polymorphisms(SNPs)are associated with RSA susceptibility, which may become a tool for monitoring RSA in the future. The integrin β3 subunit, encoded by the gene called ITGB3,is a chain protein highly expressed on the surface of platelets and embryonic tissues. It has been found that down-regulation of expression of the integrin β3 subunit is associated with RSA.We found that the expression of ITGB3 carrying rs2317384 AA type is down-regulated through database analysis, but the relationship between this SNP and RSA susceptibility has not been reported. In this study,we intend to explore the relationship between the rs2317384 from ITGB3 and RSA in women of childbearing age in southern China.MethodsIn this study,we recruited 593 RSA patients and 979 healthy controls on the basis of a case-control study, collected blood samples from them. The rs2317384 genotype of ITGB3 will be performed. We will use chi-square test and logistic regression analysis to analyze the odds ratios(ORs),95% confidence intervals(CIs)and corresponding p values between rs2317384 genotypes and RSA risk between two groups. At the same time, using age and number of miscarriages as stratified factors,we stratified the risk of RSA in specific genotypes and two groups of people to analyze the odds ratios(ORs),95% confidence intervals(CIs)and corresponding P values.ResultsIn this study, there was no significant difference in mean age between the cases and control groups(P>0.05);There was a statistically significant difference in the number of miscarriages between the cases and control groups(P<0.01);The ITGB3rs2317384 genotypes distribution conforms to Hardy-Weinberg’s law(P> 0.05).Our data analysis showed that there was no evidence that the polymorphism rs2317384 distribution of ITGB3 was associated with the risk of recurrent spontaneous abortion(GA vs AA: OR=1.086,95%CI=0.846-1.394,P=0.5176;GG vs AA: OR=0.992,95%CI=0.739-1.334,P=0.9600;dominant model: OR=1.056,95%CI=0.833-1.339,P=0.6531;recessive model: OR=0.939,95%CI=0.737-1.195,P=0.6079).The stratified analysis also showed no significant associations.ConclusionsThis study suggests that the rs2317384 polymorphism of ITGB3 may not be associated with recurrent spontaneous abortion in a Southern Chinese population. |
