| Objective: To screen postmenopausal women with osteoporosis and osteoarthritis of knee joint,and use double-blind,random,control group,placebo group and single-center large-scale clinical trials to study and compare the adjuvant treatment effects of probiotics on patients,and to explore whether there are significant differences in WOMAC score,IL-1,IL-6,IL-10,IL-17,TNF-α,IFN-γ and bone mineral density before and after treatment,which means menopause.Methods: All the patients,aged from 50 to 70 years old,who were admitted to the Affiliated Hospital of Inner Mongolia Medical University,were diagnosed as postmenopausal women with osteoporosis and knee osteoarthritis.After preliminary evaluation,80 subjects were included in the study and divided into experimental group and control group.Before the intervention,WOMAC score table was filled in,venous blood and feces were collected from all subjects,and inflammatory factors and bone mineral density were detected in our hospital.The feces were sent to Inner Mongolia Agricultural University for further analysis,and the above data were taken as baseline data.Then,the first month and the third month as the treatment period,the subjects took basic drugs,probiotics or placebo at the same time,took samples and filled in WOMAC score table,then stopped taking probiotics or placebo,and the fifth month as the clinical observation period,took samples and filled in WOMAC score table to measure bone mineral density.Compare the changes of WOMAC score and inflammatory factors between and within the two groups before and after treatment,and compare the changes of bone mineral density index before and after probiotic intervention.Results: The baseline level of the selected two groups of subjects was similar,with no significant difference.The bone mineral density of experimental group and control group did not change significantly before and after treatment,but WOMAC scores of both groups decreased before and after treatment(P < 0.05),and the change of experimental group was more obvious than that of control group.After treatment,the levels of IL-2,IL-17 and IFN-γ decreased significantly(P < 0.05),while the level of IL-6 did not change significantly before and after treatment and between two groups(P > 0.05).During the observation period,the level of TNF-α in the experimental group was significantly lower than that in the control group(P <0.05).The levels of IL-4 and IL-10,two anti-inflammatory factors,decreased during the clinical observation period of 1 month and 3 months,but the experimental group was significantly higher than the control group(P < 0.05).There was no significant difference in bone mineral density between the two groups before and after treatment(P > 0.05).Conclusion: Probio-M8 combined with calcium tablets and chondroitin sulfate can actively promote the treatment of postmenopausal osteoporosis with knee osteoarthritis.It can promote drug absorption,improve gastrointestinal function,regulate intestinal flora,reduce the level of inflammatory factors,significantly increase the level of anti-inflammatory factors,relieve knee symptoms,maintain bone mineral density and improve the quality of life of patients,and provide new ideas for conservative drug treatment of postmenopausal osteoporosis with knee osteoarthritis. |
