| Objective:HPLC method was used to determine the content of the main components of different matrix Jin-huang plasters,evaluate its quality,and systematically study the dissolution,release and transdermal effect of the drugs in each matrix Jin-huang plasters,and the inhibitory effect of Jin-huang plasters and Jin-huang powder concentrate on the common pathogenic bacteria of suppurative infection in vitro.By evaluating the quality of Jin-huang plasters with different matrix and analyzing its bacteriostatic effect,it provides a solid theoretical basis for the clinical application of this medicine in treating diseases of Yang syndrome ulcer.To explore a more suitable preparation method of Jin-huang plasters,and provide basis for commercial production in the future.Methods:1.The contents of curcumin and berberine hydrochloride in different matrix Jin-huang plasters were determined by HPLC,and the drug content in each matrix was calculated to evaluate their quality.2.The dissolution rate of Jin-huang plasters in vitro medium was determined by drug dissolution apparatus.3.The release rates of Jin-huang plasters were compared by gel diffusion method.4.Transdermal absorption accumulation of Jin-huang plasters in rats was investigated by drug transdermal diffusion apparatus.5.AGAR dilution method was used to detect the inhibitory effect and minimum inhibitory concentration(MIC)of Jin-huang plasters and concentrated Jin-huang powder on common pathogenic bacteria of suppurative infection.Results:1.The solubility of Jin-huang cream,sesame oil+yellow wax and lanolin matrix was higher under chromatographic conditions,while the solubility of the other two oil matrix were relatively low.The Jin-huang cream is not only stable in the determination of drug content,but also sticky,delicate and shiny.2.In the dissolution degree test,only the peak value of berberine hydrochloride could be measured,but the cumulative dissolution amount was far from the theoretical minimum standard value,and curcumin could not appear the peak value in the corresponding position.3.In the gel diffusion experiment,the release rate of Jin-huang cream was faster than that of oil matrix,which proved that the cream matrix had better drug release effect per unit time.In the ointment matrix,the release rate of lanolin was faster than that of sesame oil+yellow wax matrix,while the release rate of the other two oily substrates was slower.4.In the in vitro transdermal test,the transmittance of berberine hydrochloride was low,while that of curcumin could not be measured under the same conditions.Although the cumulative transmission amount of berberine hydrochloride detected per unit time was not high,we calculated according to the formula that the cumulative transmission amount of berberine hydrochloride per unit area within 28h was positively correlated with the time of administration,that is to say,the cumulative transmission amount increased with the increase of administration time.5.Each matrix Jin-huang plasters had different inhibitory effect on 5 kinds of common pathogenic bacteria of suppurative infection,among which the inhibitory effect on Staphylococcus aureus and B hemolytic streptococcus is significant,but the inhibitory effect on Escherichia coli,Streptococcus pneumoniae and Pseudomonas aeruginosa is poor.The concentrated liquid of Jin-huang powder had the strongest inhibitory effect on Staphylococcus aureus,followed by B hemolytic streptococcus and pseudomonas aeruginosa.The inhibitory effect on Escherichia coli and Streptococcus pneumoniae was not obvious.The MIC of jinhuang powder concentrate to the above three strains were 0.0125g·m L-1、0.01667gm L-1and 0.1g·m L-1,respectively.Conclusion:According to the comparison of the above experimental results,the effective components of Jin-huang cream are stable,the cream is thick and delicate,and the drug release effect per unit time,transdermal accumulation test and antibacterial test are better than oil matrix.This is a very important discovery,which provides a theoretical basis for the production standard and rational use of Jin-huang plasters in laboratory,and is conducive to further clinical application and commercialization promotion. |
