| Objective:Explore the genotype distribution of methylenetetrahydrofolate reductase(MTHFR)-677 C/T(rs1801133)polymorphism、Interleukin10(IL-10)gene promoter region-592A/C(rs1800872)polymorphism in patients with hypertensive disorders in pregnancy(HDP)of Qinghai Han Group(HDP group n=140)and normal pregnant women(control group n=145).The expression levels of MTHFR、IL-10 in plasma of the two groups and the relative expression levels of MTHFR and IL-10mrna in placental tissues of the two groups were detected to determine whether there were statistical differences.Explore the risk genes and pathogenesis of Hypertensive disorders in pregnancy of Qinghai Han Group.Methods:Using DNA extracted from blood as template,PCR-RFLP method and agarose gel electrophoresis were used to detect polymorphism typing of MTHFR-677C/T、IL-10-592A/C in the two groups,and sequencing verification was performed.Plasma MTHFR IL-10 levels were detected by ELISA.The relative expression levels of MTHFR and IL-10 mrna in the placental tissues of the two groups were detected by RT-q PCR.The above data and general clinical data were analyzed by corresponding statistical methods.Results:1.Three genotypes of MTHFR-677C/T(TT CT CC)and three genotypes of IL-10-592A/C(AA AC CC)were detected in plasma DNA of both groups.2.The genotype distribution of MTHFR-677C/T polymorphism was significantly different between the two groups(P<0.05),and the CC genotype frequency of HDP group was lower than that of control group,and the CT genotype frequency of HDP group was higher than that of control group.The C and T allele frequency of MTHFR-677C/T polymorphism was significantly different between the two groups(P<0.05),and the T allele frequency in HDP group was higher than that in control group(χ~2=5.568,P<0.05).3.The genotype distribution of IL-10-592A/C polymorphism was significantly different between the two groups(P<0.05),and the AA genotype frequency of HDP group was higher than that of control group,while the CC genotype frequency of HDP group was lower than that of control group.The distribution of A and C allele frequency of IL-10-592A/C polymorphism was different between the two groups(P<0.05),and the frequency of A allele in HDP group was higher than that in control group(χ~2=8.079,P<0.05).4.The distribution of MTHFR and IL-10 genotypes were in line with Hardy-Weinberg balance(P>0.05).5.There was no significant difference in plasma MTHFR level between the two groups(P>0.05),but there was significant difference in plasma IL-10 level between the two groups.The plasma IL-10 level in HDP group was lower than that in control group(P<0.01).6.There was no significant difference in the relative expression level of MTHFR m RNA in the placenta of the two groups(P>0.05),and there was significant difference in the relative expression level of IL-10 m RNA in the placenta of the two groups(P<0.01).The relative expression level of IL-10 m RNA in the placenta of HDP group was lower than the control groupConclusion:1.The polymorphism of MTHFR-677C/T and IL-10-592A/C is associated with the occurrence of hypertensive disorders of pregnancy in Qinghai Han Group,and the T allele in MTHFR-677C/T is the susceptibility gene of HDP,and the A allele in IL-10-592A/C is the susceptibility gene of HDP.The expression of IL-10 in plasma and placental tissue may be related to the occurrence of gestational hypertension in Qinghai Han people,while the expression of MTHFR in plasma and placental tissue may not be related to the occurrence of gestational hypertension in Qinghai Han people.2.The A allele of IL-10-592A/C polymorphism may be involved in genetic susceptibility to HDP by regulating the expression of IL-10,which is of certain value for the diagnosis of HDP. |
