Dendrobium Sw.is a rare perennial epiphytic plant of Orchidaceae(Orchidaceae)with high medicinal value.Three kinds of dendrobium with high alkaloid content:D.chrysanthum,D.parishii,D.primulinum stems,leaves and flowers of D.primulinum were studied.The distribution characteristics of the alkaloids of the three kinds of dendrobium,and the composition and content of the alkaloids were excavated.At the same time,the similarities and differences of the alkaloid composition and content of the three Dendrobium species were compared.In-depth study of the material basis of the anti-tumor activity of D.primulinum.Taking human breast cancer cell line MDA-MB-231 and human lung cancer cancer cell line A547 as the research objects,four effective polar solvents were used to extract effective components from the stem of D.primulinum,and the anticancer activity of D.primulinum was analyzed.The conclusions of this study are as follows:(1)UPLC-Q-TOF-MS/MS technology has been used to analyze and identify a total of 13alkaloid active ingredients from three types of dendrobium,Hygrine(Ⅰ),Dendroprimine(Ⅱ),Shihunine(Ⅲ),4-Hydroxy-dendroxine(Ⅳ),Nobiline(Ⅴ),N-p-feruloyl-tyramime(Ⅵ),Findlayine D(Ⅶ),N-p-cinnamoyl-tyramine(Ⅷ),cis-Dendrochrysine(Ⅸ),Dendrobine(Ⅹ),Pierardine(Ⅺ),Credidamine(Ⅻ),and Dendrochrysanine(ⅩⅢ),including 4 sesquiterpene alkaloids and 5 pyrrolidinealkaloids,2 octahydroindolizine alkaloids,2 amides alkaloids.4 alkaloids were found in D.primulinum for the first time,10 alkaloids were found in D.parishii for the first time,and 5 alkaloids were found in D.chrysanthum for the first time.(2)The alkaloids in the stems and leaves of D.primulinum have been analyzed and identified,and 6 alkaloid components have been isolated and identified,Hygrine(Ⅰ),Dendroprimine(Ⅱ),4-Hydroxy-dendroxine(Ⅳ),Nobiline(Ⅴ),N-p-cinnamoyl-tyramine(Ⅷ)and Dendrobine(Ⅹ).(3)The alkaloids in the stems and leaves of D.parishii have been analyzed and identified,and 10 alkaloid components have been isolated and identified,Hygrine(Ⅰ),Dendroprimine(Ⅱ),4-Hydroxy-dendroxine(Ⅳ),Nobiline(Ⅴ),N-p-feruloyl-tyramime(Ⅵ),Findlayine D(Ⅶ),N-p-cinnamoyl-tyramine(Ⅷ),cis-Dendrochrysine(Ⅸ),Dendrobine(Ⅹ)and Credidamine(Ⅻ).(4)The alkaloids in the stems.leaves of D.chrysanthum have been analyzed and identified,and 8 alkaloid components have been isolated and identified,Hygrine(Ⅰ),Dendroprimine(Ⅱ),Shihunine(Ⅲ),Nobiline(Ⅴ),cis-Dendrochrysine(Ⅸ),Dendrobine(Ⅹ),Pierardine(Ⅺ)and Dendrochrysanine(ⅩⅢ).(5)The alkaloids of the three types of dendrobium have been compared and analyzed.There are four common components of the three types of dendrobium,including Hygrine(Ⅰ),Dendroprimine(Ⅱ),Nobiline(Ⅴ)and Dendrobine(Ⅹ).cis-Dendrochrysine is shared by D.parishii and D.chrysanthum,4-Hydroxy-dendroxine and N-p-feruloyl-tyramime are shared by D.primulinum and D.chrysanthum.Amide alkaloids were only detected in D.chrysanthum.Tetrahydropyrrole alkaloids are the main alkaloids in D.chrysanthum,and the content of cis-dendrochrysine is the highest,reaching 3137.73μg·g-1.D.primulinum is dominated by octahydroindolizine alkaloids,and the content of dendroprimine is the highest,reaching 495.278μg·g-1.Sesquiterpene alkaloids are the dominant alkaloids in D.parishii,and the content of dendrobine is the highest,reaching 90.095μg·g-1.(6)The cell activity was measured by CCK-8,and paclitaxel was used as a positive control.The results showed that the 4 parts of the stem of D.primulinum had an inhibitory effect on the proliferation rate of human lung cancer cells and human breast cancer cells,and its inhibitory ability increased with time.And the concentration of the drug is increased,and it has the characteristics of rapid drug effect at low doses.The ethyl acetate part was treated on breast cancer cells at 2.00 mg·ml-1 for 72 h to reach the highest inhibition rate of 91%.Petroleum ether was treated with 2.00 mg·ml-1 of breast cancer cells for 72 h to reach the highest inhibition rate of 94%.
