Valeriana officinalis L.s.l.is a genus of plants in the family Valeriana,its roots and rhizomes are used in medicine,Its effects include antioxidant,anticonvulsant,antidepressant,sedative and sleep aid.In its natural environment,V.officinalis grows slowly,has a low reproductive coefficient and is in short supply,moreover the harvest period has an important impact on the content of its active ingredients.At present,with the development of tissue culture technology,adventitious root culture has become one of the effective ways to obtain active ingredients for medicinal plants.Iridoid and volatile oils are the main chemical components of V.officinalis,has antioxidant and tranquilizing and other pharmacological effects.This study was based on the optimization of the extraction conditions of volatile oil of V officinalis,the volatile oil composition and content of V.officinalis root and tissue culture grown in the field harvested in different months were analyzed by gas chromatography-mass spectrometer(GC-MS),at the same time,the cyclic ether terpene content and antioxidant activity of cyclic ether terpenes were compared by high performance liquid chromatography(HPLC)and microplate reader,respectively,provide a theoretical basis for determining and solving the problem of insufficient raw materials during the harvest period of V.officinalis.The findings are as follows:1.The best extraction conditions for V officinalis volatile oil obtained by the optimization of the response surface are:liquid-to-material ratio 26.8:1 mL·g-1,extraction temperature 70℃,extraction time 4.2 h,under this process condition,the extraction rate of adventitious root volatile oil in tissue culture of V officinalis was(3.11±0.09)%,the month with the highest extraction rate of cultivated roots is July,the extraction rate was(3.36±0.04%).2.A total of 40 compounds were detected in the volatile oil of the V officinalis root harvested in April,one of the important components of valerian volatile oil detected in it,bornyl acetate,and the only phenol detected at the same time,cis-isoeugenol,was detected.,and the relative percentage content of acevalepotriate and valepotriate is higher than that of other samples.The most diverse constituents detected in the volatile oil of V officinalis root harvested in July,there are 52 species in total,the types and contents of esters and alcohols are more numerous.46 compounds were detected in tissue cultured adventitious root volatile oils,thereinto,there are many types and contents of alcohols,alkenes and ester compounds.3.In the V.officinalis cultivation root,the highest content of valerenic acid harvested in September was 4.101 mg·g-1.The content of acetyl acevalepotriate and valepotriate harvested in April was the highest in 0.454 mg·g-1 and 11.233 mg·g-1,The valerenic acid content of the adventitious roots in tissue culture was 4.158 mg·g-1,higher than V officinalis cultivation roots.4.V officinalis volatile oil has a certain scavenging ability to DPPH,ABTS+,hydroxyl radicals,the cultivation root removal effect harvested in July is the best,the second is the adventitious roots of tissue culture,but all weaker than Vc.Therefore,it is believed that the demand is different,and the best harvest period for V officinalis roots planted in the field is also different,the best harvest period for volatile oils is July,the best harvest time for valerenic acid is September,the best harvest period for acevalepotriate and valepotriate is April.The valerenic acid content of tissue cultured adventitious roots is higher than that of valerian cultivation roots,the volatile oil extraction rate and volatile oil type are second only to July,the main chemical composition of the adventitious roots cultivated in tissue is not much different from the field cultivated valerian roots,and the cultivation cycle is short,efficient and stable,which can replace the valerian cultivation roots.
