Correlation Between SOD,NO,MDA,GSH-PX In Serum And Anxiety Or Depression In Primary Sjogren’s Syndrome

Objective The objective of this study was to observe serum oxidative stress indexes in patients with primary Sjogren’s syndrome(pSS):Superoxide Dismutase(SOD),Nitric Oxide(NO),Malondialdehyde(MDA),Glutathione peroxidase(GSH-PX),discussed the correlation between which and pSS patients with anxiety or depression,in order to provide a theoretical basis for the construction of a prediction model of pSS patients with anxiety or depression.Methods A total of 87 patients with pSS patients were included in this study.6ml venous blood was collected from patients with fasting 10-12 hours.The Hamilton Anxiety Scale(HAMA)and Hamilton Depression Scale(HAMD)were evaluated and grouped within 24 hours.At the same time,collected the basic information,and the clinical manifestations of dry mouth,dry eyes,interstitial lung disease,joint pain,and rash of the patients,and clinical laboratory indicators such as blood routine,immunoglobulin G,hypersensitive C-reactive protein,sedimentation rate within 2 weeks were recorded,as well as physical examination and disease activity index(ESSDAI)evaluation of Sjogren’s syndrome.The serum levels of SOD,NO,MDA and GSH-PX were detected to explore the correlation between these four indexes and anxiety or depression associated with pSS.Results A total of 59(67.82%)pSS patients without anxiety and depression,28 patients(32.18%)pSS patients with anxiety or depression was included this study,which was including 19 patients(21.84%)only with anxiety,9 patients(10.34%)with anxiety and depression.Comparing pSS patients with only anxiety with pSS patients without anxiety and depression,there were statistically significant differences in eye dry(χ2=4.832,P=0.028),serum MDA level(z=2.753,P=0.006)and lymphocyte count(z=2.245,P=0.025)between the two groups.There were no significant differences in serum SOD level,serum NO level,serum GSH-PX level,dry mouth,interstitial lung disease,joint pain,rash,neutrophil count,hemoglobin,platelet count,immunoglobulin G,hypersensitive C-reactive protein,erythrocyte sedimentation rate and ESSDAI.There was no statistical difference in clinical data between pSS group with anxiety and depression and pSS group without anxiety and depression.In the univariate logistic regression analysis of pSS patients group with only anxiety and pSS patients group without anxiety and depression,there were statistically significant differences in serum MDA level(OR=1.713,95%CI 1.172-2.503,P=0.005),lymphocyte count(L)(OR=0.296,95%CI 0.92-0.859,P=0.025),dry eyes(OR=5.054,95%CI 1.065-23.986,P=0.041).After multivariate logistic regression,serum MDA level(OR=1.750,95%CI 1.142-2.680,P=0.010)and lymphocyte count(L)(OR=0.216,95%CI 0.059-0.790,P=0.024)were statistically different in this groups.In the study of pSS patients with only anxiety,the receiver operating characteristic curve(ROC curve)analysis of serum MDA level indicated that AUC=0.711,95%CI 0.5699-0.853,p=0.006,cut-off value was 5.206 nmol/ml,specificity was 61%,sensitivity was 84.2%.In univariate logistic regression analysis,the serum MDA level(OR=1.660,95%CI 1.029-2.677,P=0.038)was significantly different between the pSS patients with anxiety and depression and the pSS patients without anxiety and depression.After multivariate logistic regression,the serum MDA level(OR=1.804,95%CI 1.098-2.960,P=0.020)was significantly different,but receiver operating characteristic curve(ROC curve)analysis of serum MDA level showed no predictive significance.Spearman correlation analysis showed that serum MDA level was positively correlated with anxiety(r=0.314,P=0.003),very weakly positively correlated with depression score(r=0.250,P=0.020)and serum SOD level(r=0.226,P=0.035).Conclusion This study supports the involvement of oxidative stress in pSS patients accompanied by anxiety and depression.Serum MDA levels increased,lipid peroxidation elevated in pSS patients with only anxiety.Serum MDA level was positively correlated with anxiety,which had a certain predictive value,but had no significant correlation with anxiety score.
