Background and ObjectivesThe latest cancer research statistics show that gastric cancer ranks the 3rd in incidence and mortality among all cancers in China.Since patients with gastric cancer often lack specific symptoms in the early stage,they are already in the late stage when they are detected,and patients in the late stage have poor therapeutic effect,so it is characterized by poor prognosis and high mortality.Studies have shown that collagen deposition is closely related to the development and invasion and metastasis of malignant tumors,and prolyl 4-hydroxylase subunit Alpha 3(P4HA3)is an important catalase involved in collagen synthesis,which has been found to play an important role in the development and prognosis of colorectal,lung,breast,endometrial and other malignant tumors.Therefore,this study aimed to investigate the expression of P4HA3 in gastric cancer and analyze the correlation between P4HA3 and clinicopathological factors and survival prognosis of gastric cancer patients,so as to provide new ideas for the early diagnosis and treatment and prognosis assessment of gastric cancer patients.Methods1.to analyze the expression differences of P4HA3 in cancer tissues and normal tissues adjacent to cancer in gastric cancer patients by UALCAN database and merging 3 GEO gastric cancer datasets from GPL570,and the expression differences of P4HA3 in different clinicopathological specific factors in gastric cancer patients from TCGA database;2.using Kaplan-Meier plotter online database,the median value of P4HA3 mRNA was divided into high and low expression groups to compare the differences between high and low P4HA3 expression groups in the survival prognosis of gastric cancer patients;3.In this experiment,the cancer tissues and corresponding wax blocks of normal paracancer tissues of 107 gastric cancer patients who underwent radical gastric cancer surgery in the General Surgery Department of the Affiliated Hospital of Yangzhou University were collected,and the differences in the expression of P4HA3 protein in gastric cancer tissues and normal paracancer tissues,as well as the differences in the expression of different clinicopathological factors in gastric cancer tissues were detected by immunohistochemical methods after making sections,and finally the survival prognosis was comprehensively analyzed by follow-up.4.to perform a comprehensive analysis;4.SPSS 22.0 software was used for statistical analysis and data processing.Data conforming to normal distribution were analyzed by t-test,and the results were expressed as(mean±standard deviation),and the chi-square test was used for counting data between groups,with the test level α=0.05.(1)Kaplan-Meier Plotter online database was used for survival analysis;(2)The survival prognosis of 107 gastric cancer patients at follow-up in this experiment was plotted using Kaplan-Meier method.Meier method to draw survival curves,and the Log-rank test was used for survival comparison.Differences were considered statistically significant when P<0.05.Results1.Expression of P4HA3 in gastric cancer tissues and normal tissues adjacent to cancer1.1 In both TCGA database and GEO database,the results showed that the differences in P4HA3 gene expression in gastric cancer tissues and paraneoplastic normal tissues were statistically significant(P<0.05).1.2 In 107 clinicopathological wax block specimens of this experiment,the results also showed that the expression of P4HA3 protein in gastric cancer cancer tissues and paraneoplastic normal tissues was higher in gastric cancer patients’ cancer tissues than in their paraneoplastic normal tissues,and the difference was statistically significant(χ2=4.935,P<0.05).2.The relationship between P4HA3 expression and clinicopathological factors in gastric cancer patients2.1 Comparison of P4HA3 gene expression in the TCGA database among different clinicopathological factors in gastric cancer patientsThe expression of P4HA3 gene in TCGA database in different gastric cancer patients was analyzed using UALCAN database,and the results showed that the expression of P4HA3 was related to the age,pathological differentiation degree and clinical stage of gastric cancer patients,and the difference was statistically significant(P<0.05).2.2 Comparison of P4HA3 protein expression in different clinicopathological factors in 107 clinical gastric cancer patients in this experimentP4HA3 protein expression was related to tumor diameter size,pathological differentiation degree,TNM stage,and infiltration depth T in gastric cancer patients(P<0.05);but not to patient gender,age,lymph node metastasis N,and distant metastasis M(P>0.05).3.The relationship between P4HA3 expression and prognosis of gastric cancer patients3.1 Kaplan-Meier plotter online databaseKaplan-Meier plotter online database analysis revealed that the median overall survival time,median first progression survival time and post-progression survival time in the high P4HA3 expression group were significantly shorter than those in the low expression group,and the differences were statistically significant(P<0.01);3.2 107 cases of clinical gastric cancer patients in this experimentThe Kaplan-Meier method was applied to the survival prognosis data of 107 clinical gastric cancer patients followed up in this experiment to plot the survival curves of gastric cancer patients,and the results showed that the cumulative survival time of the P4HA3 protein positive expression group was significantly shorter than that of the negative expression group,and the difference was statistically significant(χ2=14.57,P<0.001).Conclusion1.The expression of P4HA3 in cancer tissues of gastric cancer patients was significantly higher than that in normal tissues adjacent to the cancer at both gene level and protein level.2.The expression of P4HA3 gene was related to the age,pathological differentiation degree and clinical stage of gastric cancer patients;the expression of P4HA3 protein was related to the size of tumor diameter,pathological differentiation degree,TNM stage and infiltration depth T of gastric cancer patients.3.Patients with high expression of P4HA3 had poorer prognosis than those with low expression.4.P4HA3 is associated with the development and poor prognosis of gastric cancer,and is likely to be a new potential target for the treatment of gastric cancer as well as a new biomarker for assessing the prognosis of gastric cancer. |
