| Objective:Based on the theory of "lung and large intestine are exterior and interior",to explore the immune correlation between Xuanbai Chengqi Decoction and rats with heat-asthma syndrome,to investigate the action mode of Xuanbai Chengqi Decoction on related immune factors in rats with heat-asthma syndrome,to explore the possible mechanism of its regulation of immune imbalance in rats with heat-asthma syndrome,and to explore the therapeutic advantages of Xuanbai Chengqi Decoction by comparing it with Dingchuan Decoction and dexamethasone,so as to provide basis for clinical asthma classification and treatment.Methods:The method of combining theoretical research with experimental research is adopted.1.Theoretical research:Through literature analysis and theoretical research,this paper discusses the theoretical connotation and extension of "lung and large intestine are exterior and interior",summarizes the physiological characteristics of lung and large intestine and the correlation between them,and discusses the possible mechanism of treating rats with heat asthma by "treating lung and intestine simultaneously".2.Experimental study:Fifty rats were randomly divided into Group A(blank group),Group B(model group),Group C(Dingchuan Decoction Group),Group D(Xuanbai Chengqi Decoction Group)and Group E(dexamethasone Group),with 10 rats in each group.The rat model of asthma was established by sensitization with ovalbumin(OVA)and aluminum hydroxide,and the rat model of heat asthma syndrome was established by applying phlegm-heat factor.After the model was established successfully,the rats were intervened by Dingchuan Decoction,Xuanbai Chengqi Decoction and dexamethasone.The body weight,anal temperature and metabolism of rats were statistically analyzed by general data analysis,the lung function of rats was detected by lung function test,the pathological changes of lung and intestine were observed by pathological sections,and the concentrations of related factors affecting immune balance in alveolar lavage fluid(BALF)and lung and intestine tissues were detected by enzyme-linked immunosorbent assay(ELISA),so as to observe the mechanism of Xuanbai Chengqi Decoction in relieving immune imbalance of asthma.Results:Through the discussion of the theory of "lung and large intestine are exterior and interior",it is explained that there is an inseparable relationship between lung and large intestine in physiology and pathology.The imbalance of qi activity in lung will lead to the disorder of intestinal environment,and the imbalance of qi activity in intestinal tract will also aggravate the symptoms of lung diseases,resulting in pathological changes of "lung disease and intestine" and "intestine disease and lung".Through the study of the correlation between "lung and large intestine are exterior and interior" and Xuanbai Chengqi Decoction and immune balance,it is explained that Xuanbai Chengqi Decoction can be used to treat asthma with heat and asthma.The experimental results showed that asthma symptoms,anal temperature increased significantly(P<0.01),daily food intake and water intake decreased significantly(P<0.01),stool volume decreased significantly(P<0.05),airway resistance increased significantly(P<0.01)with the increase of acetylcholine concentration,inflammatory infiltration in lung and intestine tissues.ELISA showed that the levels of IL-5,IL-6 and IL-13 of type 2 helper T lymphocytes(Th2)in BALF of model rats increased significantly(P<0.01),the levels of IL-17 increased significantly(P<0.01),and the levels of IL-10 of regulatory T lymphocytes(Treg)decreased significantly(P<0.01);After the treatment,the asthma symptoms of rats were relieved,the anal temperature decreased significantly(P<0.01),the daily food intake and water intake increased significantly(P<0.01),the stool volume increased significantly(P<0.01),the airway resistance of rats decreased significantly compared with the model group(P<0.01),the pathological sections showed that the lung and intestine tissues tended to be intact,and the inflammatory infiltration decreased.ELISA results showed that the levels of Th2 cytokines IL-5,IL-6 and IL-13 in BALF decreased significantly(P<0.01),Th17 cytokines IL-17 decreased significantly(P<0.01),Treg cytokines IL-10 increased significantly(P<0.01),IL-25,TSLP and IL-33 decreased significantly(P<0.01).Conclusion:In the theoretical part,through combing the genesis and theoretical origin of the theory of "lung and large intestine are exterior-interior",the scientificity and reliability of the theory of "lung and large intestine are exterior-interior" are further clarified,and it can be used as the theoretical basis of "lung-intestine disease" in clinical practice;The experimental part shows that the inflammatory factors in the lungs of rats with heat asthma may migrate to the intestinal tissue through the homing behavior of inherent lymphocytes,causing intestinal diseases.Therefore,the application of Xuanbai Chengqi Decoction,the representative of the theory of "lung and large intestine are exterior and interior",can achieve the purpose of "treating lung and intestine simultaneously".Its specific mechanism may be to inhibit the activation of ILC2s by inhibiting the secretion of IL-25,IL-33 and TSLP in lung and intestine tissues,and reduce the secretion of related cytokines IL-5,IL-6 and IL-13,thus reducing the differentiation of ThO to Th2,thus regulating Thl/Th2 immune balance.In addition,the experimental results also show that Xuanbai Chengqi Decoction can inhibit the secretion of Th17 cytokine IL-17 and promote the secretion of Treg cytokine IL-10,indicating that it may also regulate the immune balance of Th17/Treg. |
