| Part oneCorrelation Analysis of Aβ,p-tau and Montreal Cognitive Assessment Scale Items——A retrospective analysis based on ADNIBackground and purpose:Alzheimer’s disease(AD)is the most common type of dementia.Aβ deposition and tau protein tangles are considered to be the main pathological features of AD.Therefore,the purpose of this study is to use the ADNI database to study the correlation between the general information of the subjects,the levels of Aβ and p-tau in the cerebrospinal fluid,and the results of the Montreal Cognitive Assessment(MoCA)and the scores of the subjects.Methods:A total of 86 subjects retrieved from the ADNI database were included in this study.Aβ and p-tau proteins in cerebrospinal fluid were used to reflect the pathological changes of AD;age,gender,APOE4 gene carrier status,education level,etc,of the subjects were included,and MoCA was used to reflect the changes in the cognitive abilities of the subjects.The intergroup analysis of Aβ and p-tau was performed to test the correlation between the MoCA total score and the scores of different MoCA subitems and the above biological markers.For all statistical analysis models,a two-sided p<0.05 was considered statistically significant.Result:1.Advanced age is a possible risk factor for cognitive impairment,and the MoCA score of the elderly population is low;2.Educational level is the protective factor of visual space and executive ability;3.Aβ and p-tau protein in cerebrospinal fluid are risk factors for visuospatial and executive ability;4.There is a positive correlation between the blood Aβ1-42 level and the free Aβ1-42 level in the cognitive impairment group;5.There is a positive correlation between tau level in cerebrospinal fluid of two groups of subjects and p-tau level in cerebrospinal fluid.Conclusion:Subjects with high concentrations of Aβ and p-tau protein in cerebrospinal fluid had lower scores for this item.This indicates that there is a certain correlation between AD-related biomarkers in cerebrospinal fluid and the MoCA item score,which has certain guiding significance for the early detection and early prevention of AD.Part twoCorrelation Analysis of Aβ1-42,p-tau 181 and MoCA in BloodBackground and purpose:Based on the research subjects included in the first part of the study,mainly white and black subjects,there are differences in genetic information and living habits among different races.Therefore,the purpose of this part of the study is to study the correlation between Aβ1-42 and tau protein concentration in peripheral blood and MoCA score in Chinese community population.Methods:A total of 339 middle-aged and elderly people in the community were included in this study.The pathological changes of AD were reflected by β-amyloid(βamyloid 1-42,Aβ1-42)and phosphorylated tau(p-tau)in serum;the Montreal Cognitive Assessment Scale(MoCa scale)was used to reflect Cognitive changes.The intergroup analysis of Aβ1-42 and p-tau-181 was performed to test the correlation between the MoCA total score and different MoCA sub-item scores and the above indicators.For all statistical analysis models,a two-sided p<0.05 was considered statistically significant.Result:1.Age,history of hyperlipidemia,and sleep quality are risk factors for cognitive impairment;2.The level of education is related to the scores of naming ability,attention,abstract thinking ability,language ability,delayed memory,and positioning ability.The higher the education level,the higher the scores of the above items;3.The higher the concentration of Aβ1-42 in peripheral blood,the lower the naming ability score of the subjects;4.Sleep quality is related to the level of cognitive function,thinking ability,language ability,and delayed memory scores.Subjects with poor sleep quality performed poorly in the above items.Conclusion:The results of this study provide evidence that the level of Aβ1-42 in peripheral blood plays a role in the process of cognitive function screening in community population,and point out that age,sleep quality,education level,personal disease history and community population MoCA total score and Correlation of scores.This research will help us to establish a more concise,easy and efficient Alzheimer’s disease-related cognitive dysfunction screening network,and achieve early screening,early detection,and early intervention for potential patients with Alzheimer’s disease and formulate follow-up treatment strategies. |
