Allergic rhinitis is a common nasal disease,the main symptoms of which include nasal congestion,nasal runny nose,sneezing and itching.The disease is usually caused by a variety of allergens in the air,such as pollen,dust mites,animal hair,etc.The incidence of allergic rhinitis is higher,especially in spring and autumn.Western medicine treatment of allergic rhinitis includes drug therapy and surgery.Drug therapy mainly includes antihistamines,steroid drugs,immunomodulators,antibiotics,etc.Surgical treatment includes sinus endoscopic surgery,intranasal cyst resection,nasal septum repair,etc.For the treatment of allergic rhinitis,traditional Chinese medicine also has its unique advantages and characteristics.Compared with Western medicine,Chinese medicinal materials are natural,mild,non-toxic and other characteristics,and will not cause extra burden to the patient’s body.Meanwhile,through syndrome differentiation and treatment,they can regulate the balance of qi,blood,Yin and Yang of patients,improve immunity and resistance,and have certain advantages for allergic rhinitis with complicated etiology and pathogenesis.With more than 40 years of medical experience,the instructor professor Zhou has accumulated rich experience in the clinical process,and has his own unique understanding of the etiology,pathogenesis,treatment and prescription of AR.Through the addition and reduction of the empirical prescription of Xinyishan,not only can alleviate the nasal symptoms of patients,but also can improve the Yang deficiency of patients to a certain extent,so as to achieve the purpose of promoting positive and exorcising evil.Xinyishan is a traditional Chinese medicine prescription,which has the effects of warming the middle and dispelling cold,promoting nasal orifice,relieving cough and asthma.It is widely used in clinic,especially for the treatment of cold,rhinitis,asthma,bronchitis and other diseases.This paper summarizes the experience of the tutor in treating this disease,explores the mechanism of action of the tutor Xinyishan prescription in treating this disease,and provides theoretical basis for the clinical application of AR while inheriting the tutor’s academic thoughts.Objective:Based on the method of network pharmacology,to explore the effective active ingredients,target sites and signal pathways of Xinyishan powder in the treatment of allergic rhinitis,construct biomolecular network,and analyze the mechanism of action of Xinyishan powder in the treatment of allergic rhinitis through the complex mechanism of multi-components,multi-targets and multi-pathways.Method:This research through the use of traditional Chinese medicine database and analysis platform system pharmacology,get lily magnolia in magnolia flower powder,asarum,chaff,cohosh,rhizoma ligustici wallichii,windproof,notopterygium,liquorice,angelica dehurica,also,the active ingredient,and oral bioavailability(oral bioavailability,OB)≥30%,drug-like likeness(DL)≥0.18,drug penetration of small intestinal epithelium(caco-2)≥0.4,180≤molecular weight(MW)≤500,dehydration partitioning coefficient(AlogP)<5 were selected as conditions The highly active compounds of the drugs involved in the study were obtained.Meanwhile,SMILES and 2D structures corresponding to the compounds were obtained from PubChem database,gene names were converted into Unitprot database for standardization processing,and their target sites were obtained.Cytoscape3.7.2 software was used to construct drug active ingredient target interaction network map.Results:The active components of Xinyishan powder were related to multiple targets and multiple pathways,suggesting that it may play a role in the treatment of allergic rhinitis through the action of multiple active components on multiple targets and multiple pathways.Xinyisan may act on allergic rhinitis through quercetin,kaempferol,luteolin,naringin,baicalin,β-sitosterol and other active ingredients.GO enrichment analysis showed that Xinyisan played an important role in the treatment of allergic rhinitis mainly by participating in the response to lipopolysaccharide,the response to molecules derived from bacteria,the regulation of cell-cell adhesion,the response to oxidative stress,the cell response to chemical stress,reactive oxygen metabolism and other related biological processes.Conclusion:This study analyzed the mechanism of action of Xinyishan powder in the treatment of allergic rhinitis by network pharmacology.It was found that the active components of Xinyishan powder were related to multiple targets and multiple pathways,suggesting that it may play a role in the treatment of allergic rhinitis through the action of multiple active components on multiple targets and multiple pathways.The molecular functions involved in the treatment of allergic rhinitis by Xinyi powder include cytokine activity,cytokine receptor binding,receptor ligand activity,signal receptor activator activity,transcription activator binding,DNA binding transcription factor binding,etc.Through IL-17,Toll receptor,MAPK and other pathways,JUN,CASP3,MAPK3 and other genes are acted on to play anti-inflammatory,anti-allergic and immunomodulatory roles,so as to achieve the purpose of AR treatment.This study demonstrated the characteristics of Xinyishan in the treatment of allergic rhinitis by multi-component,multi-target,multi-pathway and multi-pathway.It provides reference and new ideas for further study of its mechanism in the treatment of allergic rhinitis.
