| ObjectiveTo detect the expression changes of β2-adrenergic receptor(ADRB2)on peripheral blood neutrophils,eosinophils and basophils in patients with allergic asthma(AA),non-allergic asthma(NAA),allergic rhinitis(AR)and AR combined with AA(ARA)in acute phase,At the same time,measured the content of Tumor necrosis factor-α(TNF-α)in plasma and evaluate the lung function,in order to provide more theoretical basis for the study of the pathogenesis,diagnosis and treatment of airway inflammatory diseases.MethodIn this study,we recruited 30 patients with AA,7 patients with NAA,31 patients with AR,6 patients with ARA(the patients were all in acute phase)and17 healthy controls(HC),collected their general data(age,sex,age of onset,duration of disease).At the same time,collected their peripheral blood,and then detected the expression of ADRB2 on peripheral blood neutrophils,eosinophils and basophils of the above patients in the acute phase and HC subjects by flow cytometry,determined the level of TNF-α in plasma was by enzyme-linked immunosorbent assay(ELISA),collected data related to lung function in the lung function room,including: forced vital capacity% pred(FVC% pred)、forced expiratory volume in one second % pred(FEV1% pred)、FEV1/FVC.Result1.Flow cytometry detected the expression of ADRB2+ percentage and mean fluorescence intensity(MFI)on peripheral blood neutrophils,eosinophils and basophils in peripheral blood of patients with AA,NAA,AR,ARA and HC groups,the results show that: Compared with the HC group,the percentage of ADRB2+ neutrophils in the AA,NAA,and AR groups decreased(the difference was statistically significant),the MFI of ADRB2 on neutrophils in AA,NAA,AR and ARA groups was decreased(the difference was statistically significant).The percentage of ADRB2+eosinophils in AA,AR,and ARA groups decreased(the difference was statistically significant);the percentage of ADRB2+ basophils in AA,AR,and ARA groups decreased(the difference was statistically significant).Compared with the AA group,the percentages of ADRB2+ eosinophils and ADRB2+basophils in the AR group were decreased(the difference was statistically significant).In peripheral blood mononuclear cell population,compared with the HC group,the percentages of ADRB2+ basophils in the AA,NAA,AR,and ARA groups decreased(differences were statistically significant);compared with the ARA group,the percentages of ADRB2+ basophils in the AA,AR groups decreased(differences statistically significant).2.The contents of TNF-α in plasma of AA,NAA,AR,ARA and HC groups were measured by ELISA.The results showed that the content of TNF-α in plasma of AA,AR and ARA groups was significantly higher than that of HC group;while the NAA group had no significant difference.The content of TNF-αin AA group was higher than that in NAA group,and the difference was statistically significant.There was no significant difference in TNF-α content among AA,AR and ARA groups.3.The results of pulmonary function showed that the levels of FVC% pred,FEV1% pred,FEV1/FVC in AA,NAA,AR and ARA groups were significantly lower than those in HC group;the above lung function parameters in AA group were lower than those in AR group,but the difference was not statistically significant;compared with AA and AR groups,FVC% pred and FEV1% pred in ARA group decreased significantly,and FEV1/FVC also decreased,but the difference was not statistically significant.Conclusions1.The expression of ADRB2 on peripheral blood neutrophils and basophils in patients with AA,NAA,AR,ARA in the acute phase was decreased,which may mediate the inflammatory response process of neutrophils and basophils involved in airway inflammatory diseases.2.The expression of ADRB2 in peripheral blood eosinophils in patients with AA,AR,ARA in the acute phase was decreased,which may mediate the inflammatory response process of eosinophils involved in allergic airway inflammatory diseases.3.The content of TNF-α in plasma of patients with AA,AR and ARA increased in acute phase,no significant change in NAA patients,which suggest that TNF-α is involved in the pathogenesis of airway allergic diseases.4.The lung function of AR patients decreased in the acute phase,and the lung function of ARA patients decreased more significantly than that of asthma,patients in the acute phase,suggesting that the combination of AR may further damage the lung function of asthma patients and aggravate asthma symptoms. |
