Objective: By studying the effect of Simiao Yong’an Decoction on macrophage infiltration and GSK-3β in rats with diabetes ulcer And the expression of inflammatory factors IL-6 and IL-10.It is clear that Simiao Yong’an Decoction passes GSK-3β The effectiveness of regulating inflammatory reaction to promote the healing of diabetes ulcer,and explore the effect of Simiao Yong’an Decoction through GSK-3β Regulating inflammatory response and promoting the healing mechanism of diabetes ulcer.Methods: The rats were divided into normal group and control group by random number table.Male Wistar rats were given high fat and high sugar diet combined with small dose of streptozotocin solution(STZ)by intraperitoneal injection to establish diabetes model.The normal control group(Group C)and the diabetes group(DM group)were taken to make ulcer models,and the full-thickness skin defect wound models were prepared by back fullthickness skin resection method.They were divided into ulcer control group(CU group),diabetes ulcer group(model group DU group),diabetes ulcer-Simiao Yong’an decoction group(Simiao group-DU Simiao group),and diabetes ulcer-lithium chloride group(lithium chloride group-DU Li Cl group),with 20 rats in each group.The Simiao group was given the decoction of Simiao Yong’an decoction by gavage,the lithium chloride group was given lithium chloride aqueous solution by gavage,and the rest was given physiological saline by gavage.Observe the blood sugar of rats in each group on the 7th,14 th,21st and28 th days after diabetes ulcer modeling.Observe the change of wound healing in each group on the 3rd,7th,14 th and 21 st day after the wound modeling,and detect the change of wound macrophage infiltration by CD68 immunohistochemical staining,and detect GSK-3 in wound tissue by ELISA and Q-PCR β、Changes of IL-6,IL-10 protein and mRNA.1.After intraperitoneal injection of STZ,the blood glucose level of rats in DM group was higher than that in C group(P<0.05).2.Compared with the control group,the wound healing rate of model group,Simiao group and lithium chloride group decreased,but Simiao group was better than the other two groups3.Compared with the control group,the macrophages in the model group showed a state of slow entry and slow exit,while the infiltration of macrophages in the Simiao group was improved.4.At the 3rd,7th,14 th and 21 d after wound modeling,the expression levels of GSK-3β protein and mRNA in wound tissues of rats in all groups were ranked as follows: model group > Simiao group > lithium chloride group > control group,with differences among all groups(P<0.05).5.At the 3rd,14 th and 21 d after wound modeling,the expression levels of IL-6 protein and mRNA in wound tissue of rats in each group were in the order of model group > lithium chloride group > Simiao group > control group from high to low.On day 7,the expression levels of IL-6 protein and mRNA in tissues were ranked from high to low as follows: Model group > Simiao group > lithium chloride group > control group,there was no difference in IL-6 protein between lithium chloride group and control group(P>0.05),there was no difference in IL-6 mRNA between Simiao group and model group(P>0.05),and there were differences among other groups(P<0.05).6.At the 3rd,7th,14 th and 21 d after wound modeling,there was no significant difference in IL-10 protein level between lithium chloride group and model group(P>0.05),and IL-10 level in Simiao group was higher than that in model group but lower than that in control group(P<0.05).The mRNA expression level of IL-10 was in the order of control group > Simiao group > lithium chloride group > model group.At the 3rd and 7th days,there was no significant difference between model group and lithium chloride group(P>0.05),but there was significant difference between other groups(P<0.05).7.Compared with model group,the wound healing rate and the levels of GSK-3β,IL-6 and IL-10 in Simiao group were improved.Meanwhile,the effect of Simiao group on GSK-3β was weaker than that of lithium chloride group,and the effect on IL-10 was better than that of lithium chloride group.Conclusion: GSK-3β can delay the wound healing of diabetes ulcer rats by affecting the infiltration of macrophages and the level of inflammatory factors on the wound surface,while Simiao Yong’an Decoction can improve GSK-3β And inflammatory reaction to promote healing. |
