| The oxidative stress is caused by the imbalance between the production of Reactive oxygen species(ROS)and its antioxidant effect.Continuous oxidative stress may induce an inflammatory response,which contributes to the occurrence of a variety of human chronic diseases such as cancer,neurodegenerative diseases,cardiovascular diseases,and inflammatory diseases etc.The incidence of the Inflammatory Bowel Diseases(IBD)among people is increasing year by year and the main leading factor is the excessive oxidative stress and intestinal mucosal barrier defects.Therefore,the dietary intervention is expected to be a new approach to the prevention and treatment of related diseases.That is because many substances in diets have anti-inflammatory,antioxidant and even anticancer effects.The guts between Drosophila and human has the similar structures and functions,The characteristics of short life cycle and convenient genetic manipulation,making it an ideal model animal in studying various human diseases.Firstly,Meta analysis is used in the study to explore the effects of different extracts on the antioxidant activity of Drosophila,and further select the extracts with more significant effects on Drosophila,namely anthocyanin extracts of tangerine peel and black rice.Glutathione S-transferase(GST)is a key detoxification enzyme with anti-damage and anti-degeneration functions.In Drosophila,the Gst D1 gene encodes a GST enzyme that responds to ROS levels,So,wild-type w1118and Gst D1-GFP flies are used to investigate the effects of citrus peel and black rice anthocyanins extracts on the survival rate,climbing ability,antioxidant enzyme activities and intestinal injury protection of flies.The results concerned are as follow:1.Meta-analysis shows that different extracts can significantly prolong the life span of Drosophila,and even some extracts can improve the survival rate of the Drosophila after the oxidative stress occured.At the same time,the climbing ability and antioxidant enzyme activity of Drosophila are improved.2.The anthocyanin extracts of tangerine peel and black rice can significantly improve the survival rate of fruit flies fed by the PQ.3.The anthocyanin extracts of tangerine peel and black rice can improve the climbing ability and the antioxidant enzyme activities of the Drosophila but it works only at partial density concerned.4.The anthocyanin extracts of tangerine peel and black rice are able to clear the excessive ROS produced in the Drosophila gut due to the oxidative stress and inhibit the death of the intestinal epithelial cells to protect the integrity of the intestinal barrier. |
