Black rice extract plays a significant role in reducing blood lipid, anti-oxidation and anti-atherogenic, anti-inflammatory, regulation of the immune, prevention of cardiovascular disease and inhibiting tumor. In this paper we used Drosophila Melanogaster and Caenorhabditis elegans as model organism to study the antioxidant activity of black rice extract in vivo, and explore the molecular mechanism of its action.The Drosophila Melanogaster were fed with 0, 0.5, 1.5, 4.5 mg/mL black rice extract and the Caenorhabditis elegans were fed with 0, 70, 140, 280 μg/mL black rice extract respectively. The results were concluded via the survival test, the determination of antioxidant enzyme activity and the detection of related genes expression levels. In addition,we also did acute oxidation experiment to study the protective effects of black rice extract on the model organism.It was shown that black rice extract could prolong the lifespan of drosophila significantly, and improve the ability of crawling, enhance the activity of antioxidant enzymes containing superoxide dismutase (SOD) and catalase (CAT), then up-regulated the mRNA expression levels of CuZn-SOD、MnSOD and CAT mRNA, down-regulated the mRNA expression levels of MTH. Meanwhile, black rice extract could also enhance the resistance of flies to the acute oxidative stress effectively.Black rice extract could also make the life expectancy of Caenorhabditis elegans, and reduce the accumulation of lipofuscin. The antioxidant enzymes activity of SOD and CAT were increased and the mRNA expression levels of sir2.1、daf-16、ctl-1 and sod-3 were up-regulated, while the mRNA expression levels of age-1, daf-2 were down-regulated. At the same time, the acute oxidative stress experiment showed that black rice extract could increase the ability of resistant to acute oxidatant stress.This study showed that black rice extract can effectively delay aging of Drosophila melanogaster and Caenorhabditis elegans, enhance the activities of antioxidant enzymes,regulate the mRNA expression levels of the relevant endogenous antioxidant genes, as a result to improve the ability of scaveng free radicals and antioxidant capacity. In conclusion,black rice extract has good effects of antioxidant activity and delaying senescence.
