Objective: To observe the effect of Ruxiankang capsule on the expression level of Bcl-2 protein in breast tissue of rats with hyperplasia of mammary gland(HMG)induced by estradiol sequential progesterone.Methods:50 SPF grade female SD rats were randomly selected,and 10rats were set as blank group and 40 rats as model group.The blank group was given intramuscular injection of normal saline,and the model group was given intramuscular injection of estradiol sequential progesterone.The estradiol injection was calculated according to the body mass of 0.5 mg/kg/d,and was injected continuously for 25 days;After that,progesterone was injected sequentially,calculated according to the body mass of 5mg/Kg/d,for 5consecutive days to establish the HMG rat model.After the completion of modeling,3 rats in the blank group and 5 rats in the model group were randomly selected for model validation.After confir分钟g the success of modeling,the rats in the model group were randomly divided into the model group,the positive control group,the Ruxiankang low-dose group,the Ruxiankang medium-dose group and the Ruxiankang high-dose group,and the intervention was carried out.The blank group and model group were given distilled water by gavage,the positive control group was given Xiaoyao Pill(0.93 g/Kg/d)by gavage,and the Ruxiankang group was given low,medium and high doses of Ruxiankang capsule(0.2g/Kg/d,0.4g/Kg/d,0.8g/Kg/d)by gavage,and the gavage volume was calculated according to the body mass of10ml/Kg for 45 consecutive days.After the intervention,the body mass of rats,the diameter and height of the second pair of nipples were measured,the breast tissue of rats was extracted,the structural changes of the breast tissue of rats were observed by HE staining,and the expression level of Bcl-2 protein in the breast tissue of rats in each group was detected by IHC staining.Results:(1)Model validation: the body mass of rats in the model group was significantly lower than that in the blank group,and the diameter and height of the nipple were significantly increased.The structure and morphology of the breast tissue in the model group showed obvious pathological changes.The preparation of the rat model met the HMG model standard.(2)Body mass of rats: after intervention,compared with the blank group,the body mass of rats in the model group,positive control group,the Ruxiankang group,the Ruxiankang middle dose group and the Ruxiankang high dose group decreased significantly(P<0.01);Compared with the rats in the model group,the body mass of rats in the positive control group,the low-dose group,the middle-dose group and the high-dose group increased significantly(P<0.05);Compared with the rats in the positive control group,there was no significant difference in the body mass of the rats in the mammary gland group(P>0.05);The body mass of rats in the experimental group was slightly higher than that in the high dose group among the low,medium and high dose groups,but there was no significant difference between the groups(P>0.05).(3)Rat nipple height: after intervention,the nipple height of rats in the positive control group and the Ruxiankang group was significantly lower than that in the model group(P<0.01),and there was no significant difference between the positive group and the Ruxiankang group(P>0.05).Comparison of the nipple height of rats in the same group after modeling and intervention:the nipple height of rats in the blank group and model group after modeling and intervention was similar(P>0.05).There were significant differences in nipple height between the positive group and the mammary gland group after modeling and intervention(P<0.01).(4)Rat nipple diameter: after intervention,the nipple diameter of rats in the positive control group and the Ruxiankang group was significantly smaller than that in the model group(P<0.01),and there was no significant difference between the positive group and the Ruxiankang group(P>0.05).Comparison of the nipple height of rats in the same group after modeling and intervention:the nipple height of rats in the positive group and the mammary gland group after modeling and intervention was significantly different(P<0.01),while the nipple height of rats in the blank group and model group after modeling and intervention was similar(P>0.05).(5)The expression of Bcl-2 protein in rat breast tissue: The expression of Bcl-2 protein in the positive control group and the the Ruxiankang group was significantly lower than that in the model group(P>0.05).(6)Histopathological observation of rat mammary gland: compared with the model group,the low,medium and high dose groups and the positive group of Mammakang have significantly improved,the degree of hyperplasia can be significantly relieved,the number of acini and ducts can be reduced,the lumen area of ducts can be reduced,the secretion in the lumen is less,and the breast structure tends to be in a resting state.Among them,the breast tissue morphology of the high dose group of Mamangkang was the closest to that of the blank group,and the improvement was the most obvious.Conclusion(s): Ruxiankang capsule can improve the pathological changes of breast tissue in HMG model rats,improve the body mass of HMG rats,reduce the diameter and height of nipple,and can down-regulate the expression of Bcl-2 protein in breast tissue in a dose-effect relationship,which may be one of the mechanisms of action of Ruxiankang capsule in treating HMG.
