Objective: To observe the effect of Linggui Bafa on testicular cell apoptosis in aging model rats,and to explore its mechanism of delaying testicular aging.Methods: SD rats were randomly divided into blank,model,“Linggui Bafa”and Vitamin Egroups(n=8ineachgroup).Except the blank group,the other three groups were established by intraperitoneal injection of D-galactose for42 days.After the success of modeling,the two treatment groups began to receive acupoint moxibustion and vitamin E once daily for 28 days.The contents of SOD and MDA inserum were detected by Enzyme-linked immuno sorbentassay(ELISA).Detecting the changes in the general morphological structure of testicular tissue and the number of Leydig cells by Hematoxylinandeosin(HE)staining.Apoptosis rate of testicular cells by Terminal-deoxynucleoitidyl Transferase Mediated Dutp-biotin Nick End Labeiling(TUNEL)staining.Western blot(WB)was used to detect the expression levels of Bcl-2 and Bax in rattestis.Results:(1)Compared with the blank group,the SOD content in the serum of the model group was significantly decreased(P<0.01)and the MDA content was increased(P<0.01);The expression of BCL-2 was decreased(P<0.01),the expression of BAX was significantly increased(P<0.01);The number of testis positive nuclear expression increased significantly;The apoptosis rate of testicular spermatogenic cells increased significantly(P<0.01),the number of Leydig cells decreased significantly(P<0.01),and the damage of testicular tissue structure is serious.(2)Compared with the model group,the serum SOD content in the two treatment groups was significantly increased(P<0.01)and the MDA content was significantly decreased(P<0.01);The expression of BCL-2 in testis increased(P<0.01),the expression of BAX decreased(P<0.01);The number of testis positive nuclear expression decreased significantly;The apoptosis rate of testicular spermatogenic cells was significantly decreased(P<0.01),the number of Leydig cells was significantly increased(P<0.01)and the damage of testicular tissue structure is light.(3)There was no significant difference in serum SOD and MDA contents between the two treatment groups(P>0.05);However,the expression of BCL-2 in the testicular tissue of the Linggui Bafa group increased significantly,the expression of BAX was decreased and the apoptosis rate was decreased(P<0.05);The number of positive nuclear expression of testis in Lingguiba group was less,and the tissue structure of testis was more complete;There was no significant difference in the number of testicular Leydig cells(P>0.05).Conclusions:(1)Linggui Bafa and vitamin E can enhance the antioxidant capacity of testicular cells.(2)Linggui Bafa and vitamin E can inhibit the apoptosis of testicular cells.(3)Linggui Bafa and vitamin E can improve the internal environment of testicle tissue and restore the damaged cell structure of testicle tissue.(4)The inhibition of testicular cell apoptosis by Linggui bafa is better than that vitamin E.(5)There sults showed that the two treatment methods could inhibit apoptosis by regulating the expression of SOD,BCL-2 and BAX.The mechanism might be that they could balance them etabolism of free radical,Enhance the antioxidant capacity of the body,reduce the oxidative damage,and then inhibit testicular cell apoptosis to delay the aging of germ cells.
