| Objective: By using data mining methods such as association analysis and system cluster analysis,this paper summarizes the medication rules of traditional Chinese medicine compound in the treatment of lumbar disc herniation in the past 20 years,excavates common drug pairs,potential drug pairs and potential new prescriptions,and evolves the core combination of traditional Chinese medicine.Using network pharmacology to explore the therapeutic mechanism of core traditional Chinese medicine combination from the perspective of molecular function and biological process.Methods: Study 1: Retrieve the clinical observation literature on the treatment of lumbar disc herniation with traditional Chinese medicine compound in the database of CNKI,use the auxiliary platform of traditional Chinese medicine inheritance to count the basic information of the qualified traditional Chinese medicine compound,and establish a database for data analysis,mining the rules of medication,potential drug pairs,potential new prescriptions and core traditional Chinese medicine combinations.Study 2: Use TCMSP database to query the effective components of the core traditional Chinese medicine combination,and use Uniprot database to obtain relevant targets.Use Gene Cards,OMIM and Dis GENET databases to obtain the target points of lumbar disc herniation.Map the traditional Chinese medicine and the disease target,get the intersection target gene,upload it to the String database,and build the protein interaction network.Upload the target gene of TCM-disease intersection to DAVID database for GO and KEGG enrichment analysis.Results: Study 1: A total of 67 documents meeting the standard were included in this study,including 71 prescriptions and 143 traditional Chinese medicines,with a total of 12 traditional Chinese medicines used more than 20 times,including angelica sinensis,achyranthes bidentata,liquorice,ligusticum chuanxiong,eucommia ulmoides,white peony,mulberry parasitic,angelica,rehmannia,poria cocos,safflower and astragalus.There are 862 times of the four qi,and more than 100 times are warm,calm and cold.There are 1406 times of five flavors of traditional Chinese medicine,and more than 100 times are sweet,bitter and bitter.There are 2092 times of meridian tropism,and more than 200 times are liver,kidney,spleen and heart meridians.Set the number of support to 18(support 25%)and the confidence level to 0.6.A total of 30 groups of traditional Chinese medicine models and their frequency were obtained.Set the correlation degree to 6 and the penalty degree to 2.Through the extraction and combination function,a total of 41 association rules and the confidence between combinations are obtained.12 potential drug pairs and 6 potential new prescriptions were obtained.The core Chinese medicine combination "Eucommia ulmoides,Achyranthes bidentata,Angelica sinensis,Ligusticum chuanxiong,liquorice" was obtained.Study 2: Use TCMSP database to query,limit OB≥30%,DL≥0.18,screen and collect 24 components of Eucommia ulmoides,16 components of Achyranthes bidentata,2 components of Angelica sinensis,6 components ofLigusticum chuanxiong,88 components of Glycyrrhiza uralensis,and 4 common components.A total of 127 active ingredients of traditional Chinese medicine were obtained.After sorting out the Uniprot database,a total of 215 TCM targets were obtained.After querying Gene Cards,OMIM and Dis GENET databases,545,51 and 1 targets were obtained respectively,and 590 targets related to lumbar disc herniation were obtained after weight removal.Import the file into Cytascape 3.7.2 for drawing.The network security has 55 nodes and 804 edges.The top five core components are selected: quercetin β-Sitosterol,kaempferol,wogonin β-Carotene.The intersection target gene was imported into the String database to predict the protein-protein interaction.The results were imported into the Cytascape 3.7.2 software to draw the protein-protein interaction network,and the topology analysis of the network was carried out.The top five core targets were selected: TNF,IL-6,STAT3,JUN,AKT1,IL-1B.The intersection target genes were imported into DAVID database for GO enrichment analysis and KEGG pathway annotation analysis.After screening,151 biological processes(BP),22 cell components(CC)and 37 molecular functions(MF)were obtained.The DAVID database was used for pathway enrichment analysis to screen out the pathways for the treatment of lumbar disc herniation,including AGEs-RAGE signal pathway,IL-17 signal pathway,TNF signal pathway,MAPK signal pathway,PI3K-Akt signal pathway,TRL signal pathway,etc.Conclusion: By analyzing the common drug pairs,potential drug pairs and potential new prescriptions obtained from data mining,it can be found that the clinical application of traditional Chinese medicine in the treatment of lumbar disc herniation is mostly based on tonifying the liver and kidney,focusing on the use of drugs for warming the kidney and tonifying the liver,and regulating the spleen,promoting the absorption and transportation of the essence of the spleen and stomach,making the blood active,and paying attention to the application of traditional Chinese medicine for promoting blood circulation and removing stasis,so that blood stasis can be removed,new blood can be generated,and the qi can flow smoothly,According to the specific symptoms,they are compatible with drugs to remove wind,cold and dampness or remove dampness and heat.In clinical practice,potential drug pairs and potential new prescriptions should be carefully controlled,treated according to syndrome differentiation,and reasonably used.Through the network pharmacological research on the core traditional Chinese medicine prescription,the active ingredient quercetin β-Sitosterol,kaempferol,wogonin β-Carotene can act on TNF,IL-6,STAT3,JUN,AKT1,IL-1B and other targets,and eliminate inflammation and edema,reduce pain by regulating AGES-RAGE signal pathway,IL-17 signal pathway,TNF signal pathway,MAPK signal pathway,PI3K-Akt signal pathway,and TRL signal pathway to play the role of inhibiting inflammation,inhibiting cell apoptosis,inhibiting cell autophagy,regulating extracellular matrix degradation,and promoting nucleus pulposus cell proliferation Improve the elasticity and height of the intervertebral disc to achieve the purpose of treating lumbar disc herniation. |
