Objective: Search the literature and ancient medical records of traditional Chinese medicine treating myasthenia gravis in the past 40 years,use the software of traditional Chinese medicine inheritance assistant platform(V2.5)to conduct data mining,to explore the medication rules of traditional Chinese medicine for treating myasthenia gravis,and use the network pharmacology method to explore the pharmacological basis and mechanism of traditional Chinese medicine treating myasthenia gravis(MG).Methods: 1.Using CNKI,Wan Fang Data,VIP and Chinese Medical Code as the source and retrieval tools,respectively,search the literature on the treatment of myasthenia gravis by traditional Chinese medicine from the establishment of the database to 2022,obtain the relevant literature according to the inclusion and exclusion criteria,extract the relevant prescriptions and drugs from the literature,and establish the corresponding database.Using the platform and SPSS statistical software,the compatibility rules of the prescription were explored and summarized,and a new prescription was summarized.2.TCMSP database was used to mine the active ingredients and targets in the new prescription,and Gene Cards,OMIM and Drug Bank databases were used to screen the MG targets;Applying STRING database to build PPI network;The DAVID database was used to enrich and analyze the intersection target proteins with GO and KEGG,respectively.Results: 1.A total of 432 articles were screened,involving 455 prescriptions and 263 traditional Chinese medicines,which met the standard of Napai.The cumulative frequency of drug occurrence is 5030 times,and the top 10 Chinese medicines in the frequency of use are astragalus,atractylodes macrocephala,angelica,licorice,codonopsis pilosula,poria cocos,orange peel,cohosh,bupleurum,and yam;The spleen meridian is the highest frequency of drug meridian;The drug with the highest frequency of four gases is warm;The highest frequency of the five flavors of medicine is sweet.After the analysis of association rules,when the support is more than 30%,12 association rules of drug pairs are obtained,and the most frequent drug pair combination is Astragalus-Atractylodes macrocephala,and the core drug combinations are found to be Astragalus,Atractylodes macrocephala,Angelica sinensis,licorice,Codonopsis pilosula,and Cimicifuga.The results of cluster analysis of high-frequency drugs with frequency greater than 40 form three groups of new prescription combinations,including Buzhong Yiqi Decoction,Zuogui Decoction and Yougui Decoction,and Simiao Pill.2.A total of 159 active ingredients and 209 corresponding targets of six plant drugs in the core prescription,803 MG disease targets and 44 drug and disease intersection targets were obtained;In the active component-disease target network,the key compounds in the network,such as quercetin,kaempferol,stigmasterol,and anthocyanin,were selected according to the degree value;Ten core targets including AKT1,IL6,TP53,ALB and EGFR were screened through protein interaction network;Through GO enrichment analysis,346 biological processes(BP),33 cell components(CC)and 59 molecular functions(MF)were obtained;A total of 134 pathways were obtained by KEGG enrichment analysis.Conclusion: In the CNKI database,Wan Fang Data database,VIP database and Chinese medical code database,the idea of treating myasthenia gravis with traditional Chinese medicine oral prescription mainly follows the understanding of traditional Chinese medicine theory on flaccidity,and the high-frequency drugs selected are treated from the liver,spleen and kidney,with the main treatment principles of tonifying qi and yang,supplementing yin and yang,removing dampness and clearing heat;The treatment of myasthenia gravis with core drug combination is realized through multi-component,multi-target and multi-channel in modern pharmacological research,which preliminarily reveals the mechanism of action of traditional Chinese medicine in the treatment of myasthenia gravis and provides a theoretical basis for further research. |
