Objective: In order to implement the State Council’s Plan for the Revitalization of Zhuang medicine, establish a quality standard system(Guofa2009)No.24)and the Guangxi Zhuang Autonomous Region Food and Drug Administration’s Quality Standards for Herbs(Volume II)quality evaluation and standards research project,Christia vespertilionis. To carry out the identification of medicinal herbs, limit examination, total phenolic acid content determination, fingerprint evaluation and drug ability activity research, the initial establishment of Christia vespertilionis quality standards, for further development and utilization to provide a basis. Methods: 1.Through the collection, collation and research of the literature, the herbal textual of Christia vespertilionis of the herb is carried out;2.through the identification of original plants, morphological identification, microscopic identification and TLC identification were used for Christia vespertilionis qualitative study.3.According to the compound detection method of Chinese medicine chemistry, the pre-experiment of the chemical composition of Christia vespertilionis is carried out.4.In accordance with the methods stipulated in the four parts of the 2015 edition of the Pharmacopeia of the People’s Republic of China, the Christia vespertilionis shall be examined for water, total ash, acid insoluble ash, leachate content,etc.5.The content of total phenolic acid was determined by UV.6.at the same time, and initially established a fingerprint 7.The safety evaluation of d Christia vespertilionis was carried out through the acute toxicity experiment.8.The liver injury model of mice induced by gastric Christia vespertilionis alcohol, and then injected with D-Galactosamine(D-Galactosamine,D-Gal N)combined lipid polysaccharides(Lipopolysaccharides,LPS)in mice, was examined to investigate the liver tissue morphology of Christia vespertilionis alcohol and the expression of inflammatory factor protein levels in mouse serum and liver.ResuLts: 1.Identification of Herb of Common Christia as a Leguminosae Christian vespertilionis(Christian vespertilionis(L.f.)Bahn.f.)dry whole grass,Herb of Common Christia commonly called Herb of Common Christia, butterfly grass, bat grass;2.The original plants, traits, microscopic characteristics and physical and chemical identification characteristics of the Christian vespertilionis of the herb were found. The method of thin-layer chromatography was established, and the system was expanded with dichloromethane:methanol(16:5d),and the separation effect was good.3.Christian vespertilionis extract tested positive for organic acids, flavonoids, coumerin, sterols, sugars, tantalum, alkaloids, proteins and fats.4.A limited inspection of 10 batches of double-flying butterfly medicinal samples was carried out,and the limit standards for moisture, ash, acid insoluble ash and leachate were established: water must not exceed 12.00%,total ash must not exceed 10.00%,acid insoluble ash must not exceed 2.00%,and water-soluble leachate determination shall not be less than 8.00%.5.A method for measuring the total phenolic acid content of Christia vespertilionis was established. The total phenolic acid content of 10 batches of Christia vespertilionis was 10.33 to 16.83 mg/g,and the total flavonoid content was initially proposed to be no less than 9.00 mg/g.6.A fingerprint of the Christia vespertilionis was established, and 8 common peaks were marked, the 6th peak,as the reference peak. The similarity between 10 batches of medicinal herbs and the control fingerprint is greater than 0.96,indicating that the medicinal herbs map of different origins is more consistent and the determination method is specialized.7.The study of acute toxicity of the maximum dose of Christia vespertilionis was studied, which laid the foundation for the clinical application of Christia vespertilionis. The acute toxicity experiment found no toxic side effects related to the drug, and no mice died. All mice were examined, the appearance of each group of mice, the tissue organs were observed with the naked eye,the position, color, texture, size, etc.were not abnormal.8.Compared with the model group in the anti-inflammatory experiment, the Christia vespertilionis alcohol was significantly increased in mouse survival and reduced serum ALT,AST and liver tissue CAT,SOD,MDA,NO,GSH,GPX levels(P<0.0 5 or P<0.01),lower the expression level of liver tissue i NOS,COX-2(P<0.01),so that the pathological changes in liver tissue significantly reduced, the mechanism may be related to inhibition of i NOS,COX-2 expression. Conclusion(s): In this study, the quality standard of Christia vespertilionis of herbal textual was established, the method was fast and simple, the reproducible was good, and the results were stable and reliable, which could be used as the technical method of quality control.Christia vespertilionis alcohol is non-toxic,has a good pharmacological effect, can reduce the death rate of Galn/LPS liver-damaged mice, improve liver function, reduce inflammatory factor expression, improve the body’s oxidative stress level, its mechanism may be related to inhibiting the expression of i NOS,COX-2,for the future clinical application of Christia vespertilionis to provide some data support.Innovation point: 1.The quality standard research on Christia vespertilionis of herb provides the identification characteristics such as herbal textua, thin layer chromatography, moisture, total ash, acid insoluble ash, leachate content, total phenolic acid content,HPLC fingerprint map,etc.2.A study of the effects of the drug Christia vespertilionis alcohol on liver tissue morphology and expression of inflammatory factor protein levels in mouse serum and liver was carried out and found to have good anti-inflammatory drug activity(see Annex 2,Science and Technology Survey Report,CX2021104).
