objective:To explore the changes of autoantibody level before and after traditional treatment and belimumab treatment,and whether there is any difference between the two.Methods:The serum and clinical data of 100 patients with SLE who were treated with conventional therapy such as antimalarial drugs,glucocorticoids and immunosuppressants but not biological agents in the The First Affiliated Hospital of Suzhou University were collected.Anti-dsDNA,anti-His,anti-Nuc,anti-SSA,anti-SSB,anti-Sm,anti-PO and antinRNP in active stage(SLEDAI>9)and remission stage(6 months,SLEDAI<5)were quantitative detected by magnetic nanoparticle chemiluminescence immunoassay(NMCLIA),to explore the effect of traditional treatment on antibody level.The serum samples and clinical data of 50 SLE patients treated with belimumab regularly for the first time were collected(background treatment included routine drugs such as glucocorticoids and immunosuppressants,but not other biological agents.Belimumab10mg/kg intravenous injection:the first three doses were given once every 15 days and then once a month).According to the treatment time,the patients were divided into three groups:baseline group,3-month treatment group and 6-month treatment group.NM-CLIA was used to quantitatively detect the concentrations of anti-dsDNA,anti-His,anti-Nuc,anti-SSA,anti-SSB,anti-Sm,anti-P0 and anti-nRNP in different treatment groups,and to analyze the difference of autoantibody level during 6 months of treatment with Bailiu monoclonal antibody.And compared with the general treatment.The data were analyzed by SPSS26.0.Results:In the traditional treatment group,there was significant difference in the concentrations of anti-dsDNA,anti-His and anti-Nuc between remission and active stage of SLE,but there was no significant difference in anti-SSA,anti-SSB,anti-PO,anti-Sm and anti-nRNP.During the treatment of belimumab monoclonal antibody,there were significant differences in the concentrations of anti-dsDNA,anti-His,anti-Nuc and antinRNP among the treatment groups,but there was no significant difference in the concentrations of anti-SSA,antiSSB,anti-PO and anti-Sm among the treatment groups.Among the SLE patients who regularly used belimumab,the patients with the decreasing trend of anti-ds-DNA,anti-His,anti-Nuc,anti-nRNP and anti-PO accounted for a high proportion of the patients whose anti-SSA,anti-SSB and anti-Sm showed a downward trend.Compared with traditional therapy,the concentration ofanti-Nuc、anti-nRNP、anti-PO、anti-Sm decreased more significantly and anti-dsDNA、anti-His turned negative significantly in patients treated with belimumab monoclonal antibody.Conclusion:This study showed that the concentration of anti-dsDNA,anti-His and anti-Nuc decreased significantly in remission stage after traditional treatment,and the concentration of anti-dsDNA,anti-His,anti-Nuc and anti-nRNP decreased significantly after treatment with Bailiu monoclonal antibody.Compared with traditional treatment,belimumab monoclonal antibody treatment can significantly reduce the concentration of antibody,and a larger proportion of patients with anti-dsDNA,anti-His turned negative.
