| Objective To investigate the correlation between serum phospholipids and carotid atherosclerosis and the stability of sclerotic plaques.Methods 1 A total of 465 patients with newly diagnosed carotid atherosclerosis were selected as the plaque group.According to the results of carotid angiography,the 465 plaque group was divided into a stable plaque group of 225 cases and an unstable plaque group of 240 cases.127 cases without carotid atherosclerosis were selected as the control group,and statistical methods were used to screen the different indicators between different groups.Spearman correlation analysis was performed on the different indexes in the previous step to analyze their relationship with carotid atherosclerotic plaques.The independent risk factors of carotid atherosclerotic plaques were evaluated by binary Logistic regression model;the value of the area under the ROC curve(AUC)was evaluated by a single serum index and a combination of multiple indexes in predicting carotid atherosclerotic plaques in patients.2 Using bioinformatics technology to download phospholipid(PL)and atherosclerosis(AS)related expression profiles from the GEO database;AS plaque stability related expression profiles.Differentially expressed genes(DEGs)were screened using R software,Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis were performed using the online database DAVID 6.8,and the protein-protein interactions(PPI)of these DEGs were constructed through the online database STRING network complex.Finally,differentially expressed genes,action pathways,and key proteins both related to phospholipids and atherosclerotic plaque stability were screened.Results 1 There were statistical differences in TG,LDL-C,HDL-C,PL,and sd LDL-C between the control group and the plaque group(P<0.05).And There were statistically significant differences in PL,sd LDL-C,and Lp(a)between the stable plaque group and the unstable plaque group(P<0.05).2 Spearman correlation analysis showed that the levels of LDL-C,PL,sd LDL-C and Lp(a)were positively correlated with the progression of carotid atherosclerotic plaque,and the correlation coefficients were 0.333,0.492,0.352,and 0.213,respectively,of which Lp(a)was of weak correlated with plaque progression.3 Logistic showed that PL,sd LDL-C and Lp(a)were independent risk factors for plaque instability(P<0.05).4 The AUC values of PL,sd LDL-C and their combination in predicting the stability of CAS plaques were 0.705,0.663,and 0.724,respectively(P<0.05).5 GEO database gene statistics screened 45 differentially expressed genes(26 positively correlated genes and 19 negatively correlated genes)in PL and AS plaque stability.6KEGG enrichment analysis found that the differentially expressed genes were mainly through the regulation of cellular exosome secretion Affects AS plaque stability.7 PPI analysis identified 9 proteins(MMP9,MMP1,MMP7,MME,WNT5 B,DPP4,FZD7,IL1 B and IL1RN)closely associated with PL and AS plaque stability.Conclusion 1 Phospholipids are related to carotid atherosclerosis,and the level of phospholipids can reflect the stability of carotid atherosclerotic plaques.2 The combined detection of serum PL and sd LDL-C has a higher predictive value for judging the stability of CAS plaques than a single index.3 MMP9,MMP1,MMP7,MME,WNT5 B,DPP4,FZD7,IL1 B and IL1 RN genes and their corresponding proteins that regulate phospholipid metabolism may be new molecular targets for predicting the stability of atherosclerotic plaques.Figure10;Table4;Reference 165... |
