Objective:To investigate the relationship between recurrent spontaneous abortion(RSA)and estrogen receptor β gene Rsa Ⅰ and Alu Ⅰ gene loci polymorphism,related coagulation function,immune antibody.Methods:In this case-control study,50 patients with RSA and 50 healthy women were selected.Peripheral venous whole blood was collected from the subjects,and PCR-RFLP method was used to study the polymorphism of Rsa Ⅰ and Alu Ⅰ in ERβgene.χ2 test was used to determine whether the distribution of ERβ gene genotypes was in accordance with Hardy-Weinberg equilibrium,and the frequency distribution of genotype and allele was compared between the two groups.Logistic regression was used to calculate odds ratio(OR)and 95%confidence interval(95%CI),and the relationship between RSA and anticardiolipin antibody,antinuclear antibody,anti-lupus antibody,protein C,protein S,D-dimer,antithrombin Ⅲ,anti-β2-glycoprotein Ⅰ antibody,platelet aggregation rate activity was analyzed.The gene loci of Rsa Ⅰ and Alu Ⅰ polymorphism of ERβ gene were analyzed to investigate the clinical value of ERβ gene in RSA.Results:1,This study through the analysis of clinical data,the immune antibody and blood coagulation related inspection,think of RSA of coagulant function abnormality in the RSA group and immune function was significantly higher than that in the control group,with the increase of the number of abortions and age,coagulant function abnormality rate also increased,thrombosis,former state,the difference was statistically significant(P<0.05).2.In this study,PCR-RFLP method was used to analyze two functional gene loci of ERβ gene Rsa Ⅰ and Alu Ⅰ in RSA patients of Han Nationality in Fuzhou area.The results showed that RR,Rr and rr ratios in RSA group were 2.0%,16.0%and 82.0%respectively,and ratios in control group were:6.0%,32.0%,62.0%;The proportions of AA,Aa and aa in RSA group were 10.0%,26.0%and 64.0%,respectively,and those in control group were 8.0%,24.0%and 68.0%,respectively.rr and aa genotypes were the most frequent,and RR and AA genotypes were the least frequent.3.The results of this study showed that the distribution frequency of ERβ gene in RSA group and control group was consistent with the genetic balance test by Hardy-Weinberg genetic balance law and was representative of the population.According to χ2 test and correlation analysis,There was significant difference in the distribution of RsaⅠ loci between the two groups(χ2=6.501,P=0.039).There was significant difference in allele frequency between the two groups(χ2=7.792,P=0.005).The relative risk analysis of allele frequency showed that the OR value of R allele was 0.333,and the 95%CI was[0.151,0.738],Because OR value<1 indicates that R allele might not be a susceptibility gene locus to RSA.There was no significant difference in the distribution of AluⅠ locus between the two groups(χ2=2.212,P=0.900).There was no significant difference in allele frequency between the two groups(χ2=0.267,P=0.606).The relative risk analysis of allele frequency showed that the OR value of the A allele was 1.195,and the 95%CI was[0.608,2.349],Because OR value>1 indicates that the A allele might be risk gene locus of RSA.The distribution of antinuclear antibody was significantly higher than that of ERβ gene in RsaI group between RR+Rr and rr type(χ2=3.962,P=0.047).There was no significant difference in the distribution of anticardiolipin antibody,anti-lupus antibody and anti-β2-glycoprotein Ⅰ antibody between RR+Rr and rr type(χ2=2.415,0.195,P=0.120,0.659,0.519).Conclusion:In this study,rr and aa genotypes were the most frequent among the two functional loci of Rsa Ⅰ and Alu Ⅰ of ERβ gene in Some Han RSA patients in Fuzhou area,while RR and AA genotypes were the rarest.ERβ gene Rsa Ⅰ was correlated with RSA,and the alleles of Rsa Ⅰ enzyme segmentation site migh not be the risk locus of RSA,while the polymorphism of Alu Ⅰ locus was not correlated with RSA,but the A allele might be a susceptible gene locus Anti-nuclear antibody may be related to ERβ gene Rsa Ⅰ.in RSA patients.However,and anticardiolipin antibody,anti-lupus antibodies and anti-β 2 glycoprotein Ⅰ antibodies may not be associated to ERβ gene Rsa Ⅰ in RSA patients.
