| ObjectiveAccording to previous data,lung cancer is the highest incidence of cancer,but according to statistics in 2016-2020,the number of global breast cancer from 2.2 million cases,increased to 2.26 million cases,compound growth rate of 3%,in the world health organization international cancer research center released 2020 data we know that the number of breast cancer has exceeded the number of lung cancer,breast cancer has become the most common human malignant tumor.It is expected that the cases will reach or exceed 2.47 million in 2025,and the incidence of breast cancer ranks first among all the malignant tumors in women,far exceeding other cancer types in women.The incidence of breast cancer in women in China is 425.5/1000,000.Based on the serious adverse effects of breast cancer on women’s health,the diagnosis and treatment of this disease cannot be ignored.Although with the improvement of the level of clinical breast cancer in recent years,the progress of human social medical level,but the prognosis of breast cancer patients is not significantly improved,so to do for breast malignant tumor found as soon as possible,as soon as possible treatment and advocate postoperative precise targeted treatment,our work is still facing great challenges.There are studies that KDM5B in many human malignancies.However,according to the current studies,there are few studies on the expression of KDM5B in breast malignant tumors and the correlation between the clinical and pathological data of patients in China.In this experiment,we analyzed immunohistochemical staining of breast cancer tissue samples in the samples of breast malignant tumor,analyzed the expression of KDM5B in breast malignant tumor and the correlation between the clinical and pathological data of patients,and then systematically analyzed the significance of the expression of KDM5B in breast malignant tumor tissue.According to the clinicopathological data and follow-up data collected in this experiment,the correlation with the prognosis of breast cancer patients was further explored.The relevant content of the targeted therapy research of breast cancer should be improved to provide new therapeutic ideas and methods for the targeted treatment of breast cancer in the future.MethodsApproved by medical ethics committee affiliated to Shenyang medical college center hospital,collected in Shenyang medical college affiliated center hospital general surgery in January 2013-January 2018,60 breast cancer patients postoperative pathological wax,the selected patients all preoperative no neoadjuvant,radiotherapy,chemical therapy,the selected patients are signed informed consent,and have detailed follow-up data.Clinical data were collected systematically,including patient age,tumor size,tumor differentiation,lymph node metastasis,tumor pathological grade,TNM stage,pathological type,and prognosis.The selected experimental patients were followed up for discharge condition.The wax blocks were made into tissue sections,stained by immunohistochemistry,and the expression and distribution characteristics of KDM5B in breast cancer tissues and adjacent normal tissues were analyzed by immunohistochemistry,including the correlation between KDM5B expression and clinical and pathological data,Kaplan-Meier survival curve and Cox regression statistics to analyze the correlation between KDM5B expression and the prognosis of breast cancer patients.ResultsAccording to the experimental results,KDM5B showed highly positive expression in cytoplasm and slightly positive expression of KDM5B in breast tissue cells was significantly higher than in carcinoma tissues(P<0.05);the expression of KDM5B showed a positive correlation with lymph node metastasis,histological grade,tumor size,TNM stage,ER receptor positive(P<0.05),and had no significant correlation with age,menstrual status,histological type,PR positive and HER 2 positive of breast cancer patients(P>0.05).The results of Kaplan-Meier survival curve analysis showed that postoperative survival of patients with high KDM5B expression in breast cancer tissue was significantly shorter than that with low cancer expression(P<0.05);Cox multivariate regression analysis indicated that overexpression of KDM5B could serve as an independent prognostic factor in breast cancer patients(P<0.05).Conclusions1.KDM5B is highly expressed in breast cancer tissue cells.2.A positive correlation between KDM5B expression and lymph node metastasis,histological grade,tumor size,TNM stage and ER receptor positivity in breast cancer(P<0.05).3.KDM5B expression can be used as an independent prognostic factor in breast cancer patients after surgery(P<0.05).4 KDM5B could be a potential target for targeted therapy in breast cancer. |
