| Objective:To establish the preparation method for a long-term ABO red blood cell antigen identification reagent,to extend the storage time of red blood cells,to improve the antigen stability of clinical ABO reverse cells and antibody detection cells,and to promote standardization and standardization of detection.Methods:It was determined that 0.005% glutaraldehyde and 0.05%paraformaldehyde were the most effective chemical modification procedures for ABO reverse erythrocyte reagent.The ABO reverse erythrocyte reagent modified by 0.005% glutaraldehyde and 0.05%paraformaldehyde was developed and compared to the ABO reverse erythrocyte reagent that was not chemically treated.The performance of the ABO reverse erythrocyte reagent modified by 0.005% glutaraldehyde and 0.05% paraformaldehyde was assessed.The preparation method and preservation solution of the ABO reverse erythrocytes reagent without chemical treatment are identical to those of the commercially available reagent.Red blood cells and plasma were separated from 69 cases of anticoagulated whole blood with sodium citrate obtained from a hospital’s blood transfusion department between May 2021 and May 2022.The same plasma sample was examined by parallel cross-examination with ABO reverse erythrocytes reagent modified with 0.005% glutaraldehyde and 0.05% paraformaldehyde and ABO reverse erythrocytes reagent which had not been chemically treated.The detection indices consist of erythrocyte specificity,erythrocyte antigen strength,free hemoglobin in erythrocyte supernatant,erythrocyte appearance,erythrocyte dispersity,and the clarity of the card background when ABO reversed erythrocyte reagent is used for microcolumn gel card detection.Comparing the accuracy of ABO anti-erythrocytic reagents modified by 0.005%glutaraldehyde and 0.05% paraformaldehyde with those not chemically treated when stored at 4 degrees Celsius for 6 months,the feasibility of ABO anti-erythrocytic reagents modified by 0.005% glutaraldehyde and0.05% paraformaldehyde was determined.Results:The optimal modification types and concentrations of red blood cell reagents are determined to be 0.005% glutaraldehyde and 0.05%paraformaldehyde.The new reagent modified using 0.005%glutaraldehyde and 0.05% paraformaldehyde has a six-month shelf life.The test tube method(n=24)and microcolumn gel card(n=35)were used to determine the new reagent for long-term reverse typing of red blood cells,with an accuracy of one hundred percent.Conclusion:The antigen identification reagent for ABO reverse typing red blood cells chemically modified by 0.005% glutaraldehyde /0.05%paraformaldehyde developed in this experiment can be preserved for a long time(6 months),and the red blood cells have complete morphology,good antigen strength,stability and antibody titer,which can be used for clinical ABO reverse typing detection. |
