| Objective:To detect and compare the changes of the ratio of low density neutrophils(LDGs)in peripheral blood between children with acute asthma attack and healthy children,observe the difference of LDGs changes in children with different severity of asthma acute attack,analyze whether there is excessive activation of Neutrophil(NEU)in children with asthma acute attack,and reveal that the heterogeneity of Neu may be an important mechanism for the occurrence and development of asthma in children.Methods:1.To collect children with acute asthma attack(divided into the asthma group,n=32)and healthy children(divided into the control group,n=36),according to the severity of acute asthma attack,children in asthma group are divided into non-severe group(n=17)and severe group(n=15),and anticoagulation is taken from all subjects,and NEU and Eosinophilic(EOS)granulocyte are measured by blood cell analyzer,peripheral blood mononuclear cells(PBMCs)are isolated by density gradient centrifugation,and the proportion of LDGs is calculated by flow analysis.2.To compare the changes of LDGs levels between the asthma group and the control group,as well as between the non-severe asthma group and the severe asthma group.3.To analyze the correlation between LDGs and the severity of the disease,NEU,EOS counts in the asthma group.Results:1.Comparison of LDGs levels between the asthma group and the control group:The proportion of LDGs in the asthma group was(55.77±21.08)%,and that in the control group was(7.32±4.67)%.The level of peripheral blood LDGs in the asthma group was significantly increased,and the statistical difference was significant between the two groups(t=-13.436,P<0.001).2.Comparison of peripheral blood LDGs levels between the non-severe group and the severe group in the acute attack of asthma: The proportion of LDGs in the non-severe group was(40.75±13.37)%,and that in the severe group was(72.78±13.99)%.The proportion of LDGs in the severe group was significantly increased,and there was a significant statistical difference compared with non-critical recombination(t=-6.615,P<0.001).3.Correlationanalysis between LDGs proportion and severity,NEU,EOS in asthma group: LDGs proportion showed significant positive correlation with asthma severity(r=0.777,P < 0.001),positively correlated with NEU(r=0.589,P < 0.001),and had no correlation with EOS(r=-0.062,P > 0.05).Conclusions:1.LDGS is involved in the acute attack of asthma in children and is related to the deterioration of the disease.2.NEU is overactivated in children with acute asthma attack,and its heterogeneity may be one of the important mechanisms for the occurrence and development of childhood asthma. |
