Objective:By exploring different ways to construct LCG animal models and verifying the model,the feasibility of the model was confirmed.Transcriptome sequencing was performed on LCG tissue samples and controls at human level and animal model level respectively,and the molecular mechanism of LCG occurrence was preliminarily explored by observing the expression of transcriptome related genes.Methods:New Zealand rabbits were used to construct LCG animal models,and the animals were divided into 4 groups(normal control group,laryngeal reflux group,vocal cord injury group,laryngeal reflux + vocal cord injury group).Electronic laryngoscopy and pathological sections were used to confirm the successful construction of LCG animal models.Pathological tissues of clinical LCG patients were collected as LCG specimens,and normal laryngeal mucosa confirmed by pathology at the resection margin of total laryngectomy was used as control specimens.LCG specimens were collected after the animals were sacrificed under anesthesia,and the mucosal epithelial specimens of arytenoid cartilage in the normal control group were used as control.Transcriptome sequencing was performed on all specimens.Transcriptome sequencing data were compared and analyzed.Results:In the construction of LCG animal model,endoscopic and pathological verification were used to confirm the success of model construction,and the modeling success rate of the joint action of laryngopharyngeal reflux and vocal cord injury was significantly higher than that of the other two experimental groups.It was confirmed that the joint action of laryngopharyngeal reflux and vocal cord injury was the most appropriate LCG animal model construction scheme.A total of 4688 differential genes were identified by sequencing of human LCG and control samples.A total of 3236 differentially expressed genes were identified by sequencing of rabbit LCG and control samples.LCG differential gene expression profile was constructed.Through the enrichment analysis of human LCG and animal model rabbit LCG differential genes,it was found that the differential genes are mainly enriched in inflammatory process,immune process,signal transduction and other aspects.Il-6 dysregulation and significant enrichment of mast cells may be closely related to the occurrence of LCG.Conclusion:Laryngopharyngeal reflux and vocal cord injury can be used to construct an animal model of LCG,and the morphological and microscopic manifestations of the animal model are consistent with the clinical LCG.It may also suggests that laryngopharyngeal reflux and vocal cord injury are important risk factors for LCG.Through the transcriptome expression of LCG,it can be found that the occurrence of LCG is closely related to inflammatory response,immune response and various signal transduction.Mast cells and IL-6 may play a key role in the development of LCG.13 sheets,7 tables,95 references...
