| Purpose: Through detecting the expression of IL-33,ST2 and NF-κB in the epithelial skin tissue of Nonneoplastic epithelial disorders of vulvar(NNEDV)before and after High intensity focused ultrasound(HIFU)treatment,we discuss the relationship among IL-33,ST2,NF-κB and NNEDV.Methods: Collecting the 60 cases’ clinical medical records of nonepithelial neoplasia of vulvar and collecting 24 cases’ vulvar skin tissues before and after HIFU treatment among the 60 cases.And collecting 5cases of normal vulvar skin tissue at the same time.Using immunohistochemical and Western blot method to detect IL-33,ST2,NF-κB expression in the tissues.Result: 1.Comparison of General information: There was no statistical difference in ages between the three groups(P>0.05),and no statistical difference in course of disease between VLS groups and LSC groups(P>0.05).2.The curative effect: The curative effect was evaluated from five aspects including skin color,skin elasticity,lesion area,itching degree and tissue structure.The curative rate of VLS groups and LSC groups were 50%,51.9% and the effective rates were 50%,46.2%.The ineffective rate were 0%,1.9%.There was no statistically significant difference in efficacy between two groups(P>0.05).3.Immunohistochemical results:1)IL-33:Before HIFU treatment the mean IOD of vulvar lichen sclerosus(VLS)groups(0.041±0.002)and Vulvar lichen simplex chronicus(LSC)groups(0.041±0.004)were higher than control groups(0.033±0.008)(P=0.021,P=0.004).There was no difference among VLS groups and the LSC groups.There was no difference between VLS groups before and after treatment(0.040±0.006).There was difference between LSC groups before and after treatment(0.036±0.004)(P=0.010).After treatment there was no difference among VLS groups,LSC groups and control groups.2)ST2: Before HIFU treatment the mean IOD of VLS groups(0.038±0.003)and LSC groups(0.039±0.004)were higher than control groups’(0.032±0.004)(P=0.017,P=0.003).There was no difference between VLS groups and the LSC groups.There was no difference in VLS groups between before and after treatment(0.033±0.005).There was difference in LSC groups between before and after treatment(0.032±0.005)(P=0.001).And there was no difference among VLS groups,LSC groups and control groups.3)NF-κB: Before HIFU treatment the mean IOD of VLS groups(0.013±0.002)were lower than control groups’(0.020±0.003)(P=0.001),while LSC groups(0.030±0.005)were higher than control groups’(P=0.001).After treatment VLS groups’(0.024±0.003)was higher than before(P=0.001),and LSC groups’(0.021±0.006)was lower than before(P=0.000).There was no difference among VLS groups,LSC groups and control groups.4.Western blot results1)IL-33:Before HIFU treatment the relative expression of protein of VLS groups(0.672±0.378)were higher than control groups’(0.126±0.088)(P=0.031).LSC groups(0.742 ± 0.565)had no difference with control groups.There was no difference between VLS groups and the LSC groups.After treatment VLS groups(0.150±0.161)was lower than before(P=0.044)while LSC groups(0.107±0.133)had no change.There was no difference among VLS groups,LSC groups and control groups.2)ST2: Before HIFU treatment the relative expression of protein of VLS groups(0.756±0.318)and LSC groups(0.909±0.488)were higher than control groups’(0.294 ± 0.108)(P=0.033,P=0.049).There was no difference between VLS groups and the LSC groups.After treatment VLS groups(0.374±0.057)and LSC groups(0.334±0.148)had no changes.And there was no difference among VLS groups,LSC groups and control groups.3)NF-κB: Before HIFU treatment the relative expression of protein of VLS groups(0.164±0.029)were lower than control groups’(0.349±0.060)(P=0.005),while LSC groups(0.770±0.027)were higher than control groups(P=0.000).After treatment VLS groups(0.458±0.023)was higher than before(P=0.000),and they were higher than control groups(P=0.033).Besides,LSC groups(0.324±0.152)was lower than before(P=0.007).And there was no difference among LSC groups and control groups.Conclusion: 1.IL-33,ST2,NF-κB may be related to the occurrence and development of LSC,and their overexpression may be one of the pathogenesis of LSC.2.IL-33,ST2 may be related to the pathogenesis of VLS.3.NNEDV could be effectively treated with focused ultrasound therapy.And there was no difference between VLS groups and LSC groups in curative effect.32 Figures,13 Tables,52 reference articles. |
