Evaluation Of The Modulation Of TLR/MyD88 Signaling Pathway And The Clinical Efficacy Of Yiqi Jiedu Huayu Prescription In Patients With Ulcerative Colitis

1.ObjectiveThis study explored the clinical efficacy of Yiqi Jiedu Huayu prescription in the treatment of Ulcerative Colitis(UC)and the effect shown in the experiment of UC mouse model,and speculated the mechanism of Yiqi Jiedu Huayu prescription in the treatment of ulcerative colitis(UC)by using the observation and measurement indicators.2.MethodsIn this study,Yiqi Jiedu Huayu prescription was used to treat patients with ulcerative colitis,and its therapeutic effect was evaluated.Secondly,it was discontinued using Dextran Sulphate Sodium(DSS)to establish UC mouse models and observe the effect of Yiqi Jidu Huayu prescription on intestinal barrier related inflammatory cytokines(Interleukin-10,DSS)in UC mice.IL-10),Interleukin-23(IL-23),and whether there was an effect on the development of Toll-like receptor/Myeloid differentiation factor 88,The expression of TLR/My D88 protein was detected to determine whether there was a regulatory effect,and then the mechanism of Yiqi Jiedu Huayu formula in the effective treatment of ulcerative colitis was inferred.2.1Clinical studyThis study planned to randomly assign 80 patients who met the inclusion criteria to two treatment groups,40 each.One treatment group received Chinese herbal medicine YQJDHY combined with mesalazine granules(YQJDHY group),and the other treatment group received western medicine mesalazine granules(MSLQ group).Both treatment groups continued for one month.The differences between the two treatment groups in endoscopic mucosal recovery,clinical symptom improvement and other aspects were compared.2.2Experimental studyThis study used DSS-induced mouse UC model and randomly divided mice into four groups(blank control group,model group,YQJDHY group,MSLQ group).After continuous administration for 2 weeks,mice were sacrificed and samples were taken.ELISA method was used to measure IL-10 and IL-23 levels;Western Blot method was used to measure TLR4 and My D88 protein expression levels.3.Results3.1Clinical study: After treatment,symptoms of both groups of patients improved,but YQJDHY group had higher efficacy than MSLQ group in improving abdominal pain diarrhea,hematochezia and other aspects;The difference was statistically significant(P<0.05 or P <0.01);From clinical data point of view;The clinical remission rate of YQJDHY group was also higher than that of MSLQ group(P <0.05);The endoscopic score and Mayo score of both groups of patients improved after treatment;But YQJDHY group was better than MSLQ group(P <0.05).3.2Experimental studyCompared with model group mice;Both YQJDHY group and MSLQ group mice had improvement effects on colon gross morphology score and pathology score;The difference was significant(P <0.01);Compared with model group;IL-10 content of each treatment group mice increased significantly;IL-23 content decreased significantly(P<0.01 or P <0.05);Compared with MSLQ group;IL-10 content of YQJDHY group mice increased significantly;IL-23 content decreased significantly(P <0.01 or P <0.05).Westorn blot method detection results showed that: TLR4 and My D88 protein expression in colon tissue of blank control mice was lowest;TLR4 and My D88 protein expression in colon tissue of model mice was significantly higher than that of each administration group(P<0.05);TLR4、My D88 protein expression in colon tissue of both YQJDHY Group And MSLQ Group Mice showed an increasing trend(P <0.05)。4.ConclusionYiqi Jiedu Huayu Formula can significantly alleviate the clinical manifestations of ulcerative colitis patients,indicating that it is safe and effective,and deserves to be widely applied in clinical practice.Yiqi Jiedu Huayu Formula can alleviate various symptoms induced by DSS-induced UC mice,increase IL-10 content,decrease IL-23 content,inhibit activation of TLR4/My D88 signaling pathway,and its effect is comparable or even better than western medicine mesalazine granules.
