Objective:This article aims to analyze the quality differences between G elata Bl.f.elata×glauca.and G.elata Bl.f.elata and G.elata Bl.f.glauca S.Chow,to explore whether hybrid G.elata Bl.has the excellent characteristics of the gastrodiac,and for its subsequent food care development provides scientific basis and methods.Methods:1.Using UHPLC-QTART-MS,select the C18 chromatographic column,the flow phase is 0.1%formic acid water-acetonitrile,the gradient is eluted.31 amino acids and nucleoside activated ingredient content,combined with the main component analysis and the advantages and disadvantage distance method for comprehensive evaluation of the quality of different batches of samples.2.Samples are used to digest the method of digestion of electric heating and humidity,with 45Sc,73Ge,115In,209Bi as the internal standard,and then use ICP-MS to determine 27Al、48Ti、55Mn、57Fe、59Co、60Ni、63Cu、66Zn、71Ga、75As、82Se、85Rb、88Sr、95Mo、111Cd、133Cs、137Ba、208Pb in the sample.The two statistical methods of PCA and the minimum two-multiplied discrimination(OPLS-DA)analyzed the results and evaluated the results.3.On the basis of the single factor experiment,the response face method is adopted to inspect the impact of extracting time,extraction temperature,and material liquid comparison to extract Gastrodia’s polysaccharides,and finally obtain the best extraction process conditions;The polysaccharides content of G.elata Bl.;use the pre-column derivatives combined with Ultra-high-efficiency liquid chromatography to determine the three monosaccharides in G.elata Bl.4.ICR mice are randomly divided into blank control groups,positive phar maceutical groups,polysaccharides(G.elata Bl.f.glauca S.Chow,G.elata B l.f.elata,hybrid G.elata Bl.),water liquid group(G.elata Bl.f.glauca S.Chow,G.elata Bl.f.elata,hybrid G.elata Bl.).The impact of polysaccharide s and water liquid liquid on mice on mouse activity on mice;then use mice’s sleep rate,sleep elements of G.elata Bl.f.elata(16.82 mg/kg)>G.elata Bl.f.glauca S.Chow(9.23 mg/kg)>hybrid G.elata Bl.(4.74 mg/kg),using Pilson’s relationship numbers to combine thermal diagram of the elements of hemps and antagonistic effects in the elements of hemps.;Through PCA,the comprehensive evaluation of 21 Castle samples showed that the S14 Jinzhai G.elata Bl.f.elata was the highest.There are rare intersections with hybrid G.elata Bl.and G.elata Bl.f.elata.3.The best extraction process obtained by the response surface optimization is the extraction time of 48min,the extraction temperature is 60°C,and the material is 1:33.Under this condition,the polysaccharides obtain rate is 32.26%;polysaccharides of hybrid G.elata Bl.obtain rate is 26.41%±0.07%,polysaccharides of G.elata Bl.f.elata obtain rate is 25.14%±0.10%,and polysaccharides of G.elata Bl.f.glauca S.Chow gain rate is 15.17%±0.04%.The three types of glutinous rice polysaccharides are similar,all of which are composed of mannose,mouse sugar,glucose,semi-lactal acid,and glucose aldosilic acid.The average Moore mass ratio of the three G.elata Bl.is 1.00:2.36:2.27:4.70:25.40.4.Three glycemic groups in the threshold dose experiment can extend the sleeping duration of mice compared to the blank group,but the extension of the two G.elata Bl.extension of G.elata Bl.f.glauca S.Chow and G.elata Bl.f.elata is not significant(P>0.05).The extension of the polysaccharides group is more significant(P<0.05);the three G.elata Bl.and the blank group in the water liquid group can significantly extend the sleeping time of mice(P<0.05),and there is no significant difference between the three groups(P>0.05).The dosage group can increase the GABA content in the mouse brain tissue compared to the blank group.Except for the polysaccharides of G.elata Bl.f.glauca S.Chow group,the other groups are significantly different(P<0.05);in polysaccharides group,hybrid G.elata Bl.is significantly different from the others(P<0.05),and there is no significant difference in the water liquid group(P>0.05);the water liquid and the polysaccharide group are not significantly different than the polysaccharide group except the polysaccharide group except the polysaccharides group(P>0.05).The measured 5-HT content showed that the polysaccharides group failed to significantly increase the 5-HT concentration in the brain tissue,while the groups of the water liquid could be significantly improved,but there was no significant difference between the group(P>0.05).Conclusion:This article determines the 21 amino acids and 10 nucleoside components of the 3G.elata Bl.through UHPLC-QTART-MS technology,and successfully detects the eight types of essential amino acids of the human body.The statistical results show In the pro-generation G.elata Bl.;the use of ICP-MS technology to successfully detect 18trace elements in three kinds of G.elata Bl.,including 7 essential trace elements.There is a antagonistic or synergistic relationship between some elements,and the differences between different varieties are significantly different.On the basis of the experiment,the regression equation is established in combination with the response surface method to obtain the best extraction process of ultrasound assisting extraction of gastrobossee.Elemental acid,semi-lactal acid,and glucose composition,different varieties of Gastrodia monosaccharides different ratio;by administering mice for 2 weeks,inspecting polysaccharides of G.elata Bl.,water liquid liquid autonomous activities,and sodium pentopoly to mice The effects of falling asleep,sleep incubation and duration of sleep,and then determine the content of inhibitory neurotransmitters in the mouse brain tissue,which proves that all three types of G.elata Bl.have sedative and hypnotic medicinal effects,but there is no significant difference between the three G.elata Bl..The comparison results of the polysaccharides group and the water liquid group and the blank group indicate that both have hypnotic effects.The efficiency of the water liquid group is significantly stronger than the polysaccharide group.The concentration of inhibitory neurotransmitters.This research results provides a scientific interpretation of the study of the Jinzhai hybrid G.elata Bl.and its own family relationship.At the same time,it provides suggestions and ideas for the development of G.elata Bl.in the field of food care and resource utilization.
