Objective:The correlation between C1ql4 expression and the stage of breast cancer and other clinicopathologic indicators in breast cancer tissues was analyzed and the role of C1ql4 as a new cancer marker and therapeutic target in the occurrence and development of breast cancer was also discussed.Methods:The corresponding case data were collected through the electronic medical record system,such as age,menstruation,maximum tumor diameter,lymph node metastasis,histological grade,molecular classification,TNM stage,etc.The surgical tissues of 118 patients with breast cancer were collected from June 2021 to June 2022 in the Department of Breast surgery of the affiliated Hospital of Chengde Medical College.The expression of C1ql4 mRNA in breast cancer tissues and some adjacent non-tumor tissues was detected by Quantitative real-time PCR.The level of C1ql4 protein in 118 patients with breast cancer was detected by Western-Blotting.Results:1.Expression of C1ql4 mRNA in breast cancer and para-carcinoma tissues.The results of q RT-PCR showed that the expression of C1ql4 mRNA in breast cancer tissues[365.13(68.12,1549.56)] was significantly higher than that in normal tissues[1.84(0.37,3.00)](P<0.05).2.Relationship between C1ql4 mRNA and protein expression and clinicopathological features.qRT-PCR results showed that the expression of C1ql4 mRNA in stage Ⅲbreast cancer[3343.79(1015.00,7843.42)] was significantly higher than that in stage Ⅱ breast cancer[561.81(312.09,1083.99)](P<0.05),and the expression in stage Ⅱ breast cancer was significantly higher than that in stage Ⅰ breast cancer[45.27(22.97,102.28)](P<0.05).WB showed that the expression of C1ql4 protein in stage Ⅲ breast cancer[1.03(0.95,1.24)]was significantly higher than that in stage Ⅱ breast cancer[0.70(0.66,0.79)P< 0.05],and the expression in stage Ⅱ was significantly higher than that in stage Ⅰ breast cancer[0.47(0.34,0.53)(P<0.05)].The results showed that the expression of C1ql4 was positively correlated with TNM stage,which was statistically significant.q RT-PCR and WB showed that the expression of C1ql4 was significantly increased in lymph node metastasis group,tumor diameter larger than 2cm group and histological grade Ⅲ group.There was no significant difference between the expression of C1ql4 and age and menopause(P>0.05).3.Relationship between C1ql4 mRNA and protein expression and molecular typing of breast cancer.The results of q RT-PCR,WB showed that the expression of C1ql4 in triple negative breast cancer was significantly higher than that in other non-triple negative breast cancer.(P< 0.05).However,the results showed that there was no significant difference in the expression of C1ql4 among different non-triple negative breast cancer(P>0.05).Conclusions:1.The expression of C1ql4 in breast cancer is high,indicating that it may be related to the occurrence of breast cancer.2.Because of the positive correlation between C1ql4 and tumor size,lymph node metastasis and TNM stage,it is suggested that C1ql4 is involved in the development of breast cancer and plays an important role in evaluating the progression of breast cancer.3.The high expression of C1ql4 in breast cancer,especially in triple negative breast cancer tissues indicates that it may be a new target for the treatment of breast cancer in the future. |
