Background: Di(2-ethyl-hexyl)phthalate(DEHP)is a common plasticizer added to everyday products.DEHP promotes drosophila senescence.Maternal exposure to DEHP during pregnancy induced birth defects in offspring and increased susceptibility to disease in adulthood.However,the effect of parental exposure to DEHP on progeny aging remains unclear.Objective: To explore the effects of parental DEHP exposure on the aging of offspring drosophila and the fetal-derived mechanism.Methods: The parental toxicity of food containing 1% DEHP was evaluated by adding a bright blue method to measure the food intake of parental drosophila.HPLC-MS was used to detect the contents of DEHP metabolites MEHP,MEOHP and MEOHP in parental drosophila after eating 0,0.01%,0.1% and 1% DEHP medium for 24 h,confirming that the DEHP medium prepared in this experiment was effectively introduced into drosophila.At the same time,DEHP and its metabolites in fertilized eggs produced after parental exposure to DEHP were detected to determine whether parental drosophila exposure to DEHP directly affected the offspring.The survival of offspring drosophila exposed to 0,0.01%,0.1% and 1% DEHP were recorded,and the lifespan model of offspring exposed to DEHP was established.The number of eggs laid by parental,the number and sex ratio of emergence,the body length and weight of the third instar larvae,the pupation rate and the emergence rate of the offspring were counted after the parents were exposed to non-dose DEHP respectively,so as to discover the stages and manifestations of possible changes in the development of the offspring drosophila.The transcriptomic changes were detected in the eggs of the offspring exposed to 1%DEHP in the parents to find the changes in gene expression in the early life of the offspring drosophila.To detect the variation trend of body weight and triglyceride content in offspring at different life stages and explore the relationship between body weight,triglyceride and life span.Finally,the LAM system was used to detect the movement ability of the offspring drosophila along with its life span,and the relationship between the movement ability and the lifespan affected by DEHP was studied.Results: Food intake test showed that DEHP increased the food intake of parental male drosophila and decreased the food intake of parental female drosophila in a short period,and this effect was weakened with the extension of feeding time.HPLC-MS assay showed that parental drosophila DEHP metabolized normally after feeding,and the metabolite content increased with the increase of DEHP concentration in feeding medium.Parental exposure to DEHP shortened median and maximum lifespan of male and female offspring.DEHP reduced the number of parental spawning and limited the development of offspring in the third instar larval stage,but did not affect the pupation and emergence stage in the later stage of development.Transcriptomic results showed that carbohydrate digestion and absorption and neuroactive ligand-receptor interactions were impaired in eggs.Offspring with abnormal lipid metabolism and motor capacity appeared earlier in the parental DEHP exposure group than in the control group.Conclusion: Parental exposure to DEHP can affect metabolism and motor capacity of offspring,shorten healthy life span and promote aging by altering transcriptomic characteristics of eggs.It reflects the relationship between senescence and embryonic damage.
