| Vitiligo is a disease that affects the melanin of one’s own skin and has affected about 1% of people worldwide.The most important thing is that it is not only the best way to get the most out of your skin,but also the best way to get the most out of your skin.Fufang Honghua Buji(FHB)is an in-house preparation of Shanghai Dermatology Hospital,which has been clinically used in the treatment of vitiligo for more than 30 years without obvious adverse reactions and has been praised by many vitiligo patients,and its new drug transformation prospects are promising.Therefore,this project takes FHB as the research object,firstly,the qualified Chinese herbal medicine is screened by basal investigation,then the extraction process of water extraction and alcohol precipitation is optimized and a complete set of quality standard system is established for quality control,and finally the pharmacodynamic study is carried out as the pre-clinical research work by combining network pharmacology and molecular docking,in order to expect to develop it into a new Chinese medicine with stable quality and good efficacy.In the first chapter,we firstly investigated the basal origin of FHB by thin-layer chromatography,screened the herbs that conformed to the pharmacopoeia,and screened the origin of the herbs to provide the theoretical basis for selecting the optimal herbs for the subsequent formulation of FHB.In the second chapter,the quality standard of FHB was improved,firstly,the extraction method of water extraction and alcoholic precipitation of the original extraction process was optimized,and the content of psoralen and iso-psoralen and the yield of dry extract were used as evaluation indexes to preferably select the best water extraction and alcoholic precipitation process,and the number of times of extraction was investigated.The optimum conditions of hydroextraction and alcoholic sedimentation were finally established in combination with the production cost,and the optimum conditions were applied to the quality control of the preparation process.Then the fingerprint of FHB was established and 16 stable characteristic peaks were found.By comparing with the original herbal material,the 16 characteristic peaks appeared in the fingerprint spectrum were attributed,and three peaks were attributed to psoralen and iso-psoralen and hydroxy saffron yellow pigment A,and then the fingerprint spectrum was also examined methodologically.In addition,a thin-layer identification based on safflower,psoralen,astragalus and heshouwu was also established in this chapter.In addition the examination experiments under the relevant item of granules proved that the water content of FHB did not exceed 3%,its resting angle was less than46,its bulk density was between 0.46 and 0.49,and its solubility was stable and good in accordance with the pharmacopoeia.In Chapter 3,network pharmacology,molecular docking and animal experiments were used to evaluate the active ingredients and therapeutic targets of FHB.By intersecting the targets of FHB action with those related to vitiligo,94 targets were generated,of which RELA,STAT3 and AKT1 may be potential therapeutic targets.Then,based on the results of KEGG pathway analysis,the JAK-STAT signaling pathway may be involved in the therapeutic mechanism of FHB for vitiligo.In addition,molecular docking showed that lignocaine,quercetin and baicalein,the main components of FHB,bound to STAT3 targets most effectively and stably.For experimental validation,we established a mouse model of vitiligo and used ELISA,q RT-PCR and immunohistochemistry to confirm the therapeutic effect and molecular mechanism of FHB.The results showed that FHB could promote skin melanogenesis to treat vitiligo mice by reducing inflammation levels and inhibiting the molecular mechanism of JAK-STAT signaling pathway.Although in the preliminary quality standard control,we established hydroxy saffron yellow pigment A,psoralen and isopsoralen as the main index components,but the later network pharmacology and molecular docking and animal experiments predicted that lignocaine,baicalein and quercetin might be important components for the efficacy of FHB,in this study lignocaine,baicalein and quercetin as the index components were missing in the In this study,the quality control of lignocaine,baicalin and quercetin as the index components was missing,therefore,in the follow-up study,the quality standard of FHB should also include the quality control of lignocaine,baicalin and quercetin as the index components.This study demonstrated the molecular mechanism of anti-inflammatory and melanogenesis promotion of FHB through network pharmacological prediction combined with molecular docking and experimental validation,which provided new ideas and clues for the development and treatment of vitiligo prescriptions and provided experimental basis for clinical treatment. |
