Objective: To identify the pathogenesis and molecular targets of Echinococcus granulosus extravasation sensitization.Methods: Firstly,the Echinococcus granulosus and sensitization models of Balb/c mice were constructed,and the experimental groups were divided into anaphylactic group(ANPC),non-sensitized group(CE)and normal control group(CTRL).The spleen tissues were then taken for mi RNA sequencing,and the differential expression of mi RNA was found through bioinformatics analysis.PPI networks were then used to identify key target genes regulated by mi RNA related to hydatid sensitization.Results: There were 29 Differential expression of mi RNA(DEM)between ANPC vs CTRL and CE vs CTRL,respectively.Subsequently,four DEM’s shared between the two data sets were identified by Venn diagram analysis,while 25DEM’s sensitized to hydatid infection were specifically expressed.Based on the 25 DEM,the mi RNA-m RNA interaction was screened,and 174 potential target genes were identified.We studied GO function and KEGG pathway enrichment of 174 target genes.The results showed that PI3K-Akt signaling pathway,Fox O signaling pathway and Focal adhesion pathway were the most enriched.The analysis of interaction between PPI and MIRNA-HUB gene network showed that IL-6 was regulated by MIR-146A-5P and MIR-149-5P.IL-10 regulated by MIR-29B-3P and MIR-29C-3P was detected by RT-PCR,and compared with the control group,IL-6 and IL-10 were significantly improved in hydatid infection and sensitization group.The PI3K-Akt signaling pathway,Fox O signaling pathway and Focal adhesion signaling pathway participate in the development of hydatid sensitization.Conclusion: After cyst rupture in CE patients,mi RNAs regulating IL-6 and IL-10 are significantly increased,and IL-6 and IL-10 play an important role in cell pyro-death,apoptosis and endoplasmic reticulum stress.Therefore,the above results not only help to find the regulatory targets related to IL-10 and IL-6,but also help to study the pathological processes of pyro-death,apoptosis and endoplasmic reticulum stress in hydatid sac fluid sensitization.Thus,it can help us elucidate the mechanism of DEM in the three cell damage processes of pyro-death,apoptosis and endoplasmic reticulum stress in the allergic reaction caused by hydatid cyst fluid,so as to provide tissue and organ protection strategies for hydatid patients with severe allergic reactions,and accelerate the recovery of patients.
